Abstract: Mutant isocitrate dehydrogenase 1 (mIDH1; IDH1R132H) exhibits a gain of function mutation enabling 2-hydroxyglutarate (2HG) production. 2HG inhibits DNA and histone demethylases, inducing epigenetic reprogramming and corresponding changes to the transcriptome.
