Epigenetic regulation during melanocyte development and homeostasis

Abstract: Melanocytes originate in the neural crest as precursor cells which then migrate and proliferate to reach their destination where they differentiate into pigment-producing cells.Melanocytes not only determine the color of hair, skin and eyes but also protect against the harmful effects of UV irradiation. The establishment of the melanocyte lineage is regulated by a defined set of transcription factors and signaling pathways that direct the specific gene expression programs underpinning melanoblast specification… Show more

“…CCNB1 may regulate melanocyte proliferation through the p53 signaling pathway [ 82 , 83 ]. In melanocytes, MITF regulates DCT gene expression by forming a protein complex with EP300 [ 84 ]. ADCY10 deletion experiments demonstrated its association with the proliferation of eumelanin [ 85 ].…”

“…The role of SOX10 in the life history of adult bulbar melanocytes is also intriguing, given that the expression of this transcription factor is required for melanocyte/melanoblast commitment in utero [ 44 ]. Is SOX10 expression dispensable for some melanocytes in the adult hair bulb?…”

“…Melanoblasts originating from embryonic neural crest cells invade the epidermis from the dermis and differentiate into melanocytes. Differentiated epidermal melanocytes migrate into hair follicles and constitute hair bulb melanocytes, where mature stage IV melanosomes are transferred to surrounding keratinocytes, giving rise to melanized hair (4)(5)(6)(7)(8). Melanized hair contributes to the expression of the coat color in animals.…”

“…Hair bulbs at the bottom of the hair follicles contain keratinocytes, melanocytes, and fibroblasts (special fibroblasts called dermal papillae cells). The hair bulb melanocytes secrete mature melanosomes into surrounding keratinocytes, producing melanized hair (1,(4)(5)(6)(7)(8).…”
