Epigenetic modulator UVI5008 inhibits MRSA by interfering with bacterial gyrase

Abstract: The impact of multi-drug resistant bacterial strains on human health is reaching worrisome levels. Over 2 million people are infected by resistant bacteria, and more than 700,000 people die each year because of the continuous spread of resistant strains. The development of new antibiotics and the prudent use of existing ones to prolong their lifespan require a constant effort by drug industries and healthcare workers. The re-purposing of existing drugs for use as antimicrobial agents would streamline the devel… Show more

“…Nowadays, the growing threat of microbial resistance against traditional antibiotics in oral disease has prompted the mandatory development of alternative therapies, especially against bacterial strains showing resistance to one or more antibiotics [60,61]. In this article, silver NPs (AgNPs) were successfully prepared using hydrazine and were further modified by a coating of indolicidin, a well-known AMP.…”

“…e Gramnegative and -positive isolated bacteria in our University Hospital had shown a low rate of resistance to the fluoroquinolones tested (ciprofloxacin, moxifloxacin, and levofloxacin). Antibiotics, belonging to this class, inhibit DNA gyrase and topoisomerase IV, enzymes that are involved in DNA replication [39]. ese drugs are broad-spectrum antibiotics, providing excellent coverage against most ocular pathogens [40].…”

“…Epigenetic regulations refer to the complex and multilayered mechanisms involved in the variation of a gene expression without a change in the DNA sequence itself [ 92 ]. Epigenetics represents a pivotal mechanism of cellular and organ homeostasis, which includes various regulatory events, such as DNA methylation, histone modifications and noncoding RNAs [ 93 , 94 ].…”
