“…It's well known, analysis the capacity to form multi-cellular spheroids is a commonly used technique to evaluate the self-renewal potential of CSCs in many solid tumors [12]. In our present study, we found that the ability to form spheroids in sphere culture was markedly restricted after restoration of miR-200a expression.…”

“…SOX2 expression has been demonstrated to improve the ability of invasion and migration of tumor cells in tongue squamous cell carcinoma (TSCC) (77). Other properties of cancer cells in OSCC or HNSCC that involve the SOX2 protein include apoptosis (74,78), chemoresistance (79), metastasis and tumorigenesis (10,79).…”

“…It has been also shown that synergistic treatment of EGCG with quercetin led to a decrease of CD44+/ CD133 + prostate cells through inhibition of Vimentin, Slug, Snail, and nuclear b-catenin expression and LEF-1/TCF responsive reporter activity, affecting the CSC migration and invasion (Tang et al, 2010), and in combination with curcumin, it led to a reduction of sphere formation from breast tumor cells by inhibition of STAT3 phosphorylation and retention of STAT3-NFkB interaction (Chung & Vadgama, 2015). Moreover, in head and neck squamous cell carcinoma (HNSCC), EGCG treatment has been reported to downregulate the expression of stem cell markers, such as Oct4, Sox2, Nanog, and CD44 and to suppress HNSCC cells sphere forming capacity, augmenting their cisplatin (CP)-mediated chemosensitivity by inhibiting ABCC2 and ABCG2 transporter genes (Lee et al, 2013b). In the colorectal tumor model, EGCG induced sensitization of the 5-fluorouracil (5FU)-resistant colorectal cancer cells and spheroid-derived colorectal CSCs to 5FU treatment that was associated with suppression of Notch1, Bmi1, Suz12, and Ezh2, upregulation of self-renewal suppressive-miRNAs, miR-34a, miR-145, and miR-200c and enhancement of apoptosis and cell-cycle arrest (Toden et al, 2016).…”
