Epidermal growth factor receptor tyrosine kinase is modulated by GM3 interaction with N-linked GlcNAc termini of the receptor

Abstract: Epidermal growth factor receptor (EGFR) at membrane microdomains plays an essential role in the growth control of epidermal cells, including cancer cells derived therefrom. Ligand-dependent activation of EGFR tyrosine kinase is known to be inhibited by ganglioside GM3, but to a much lesser degree by other glycosphingolipids. However, the mechanism of the inhibitory effect of GM3 on EGFR tyrosine kinase has been ambiguous. The mechanism is now defined by binding of N-linked glycan having multiple GlcNAc termini… Show more

“…Although the GD3S-KO NSCs may accumulate GM3, which has been shown to inhibit tyrosine kinase activation by binding to EGFR (22), our findings in this study point to an alternative mechanism by which gangliosides may regulate that growth factor receptor activity.…”

“…Because binding of EGFR with GM3-coated beads has been reported by previous studies (21,22), a GSL-coated bead assay was also used to determine whether colocalization and coimmunoprecipitation of EGFR and GD3 represent a bona fide biochemical interaction. A Western blot with anti-EGFR antibody showed binding of EGFR from NSCs lysate in GD3S +/+ cells with GD3-and GM3-coated polystyrene beads, but not with Fig.…”

“…This possibility was tested by binding of CD82 to polystyrene beads coated with GSLs, using the procedure described in ref. 11. CD82 bound more strongly to beads coated with GM2-GM3 mixture than to beads coated with GM2 or GM3 alone or GM2-LacCer mixture (Fig.…”

“…They are major components of membrane microdomains known as "lipid rafts" (22) and are important players in cell signaling (23) and cell-cell interaction (24). By influencing membrane properties and/or by direct interaction with membrane proteins, gangliosides modulate the activity of many tyrosine kinase (25)(26)(27)(28) and neurotransmitter receptors (29), ion channels (30,31), and downstream cell signaling pathways. In addition, gangliosides regulate axon-myelin communication and the maintenance of myelinated axons in the adult central and peripheral nervous systems (32)(33)(34).…”
