Epidemiological Study of AGT Gene Polymorphism among Chinese Subjects with Primary Hypertension

Abstract: Background: Polymorphisms in the promoter region of the angiotensinogen (AGT) gene may affect AGT transcription and thus blood pressure. We determined the frequency of the AGT A-20C polymorphism in Chinese patients with primary hypertension. Using a molecular epidemiology approach, we also determined the relationship between primary hypertension and environmental-AGT A-20C polymorphism interactions. Methods: 912 subjects diagnosed with primary hypertension were analyzed from Shenyang Chinese Populations and the… Show more

“…The AGT A-20C gene polymorphism was investigated by polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) technique using the primers (from Eurogentec) and restriction enzyme (FastDigest EcoO109I from Thermoscientific) as previously described [9]. The PCR conditions were similar to those described by Zhang et al [13].…”

“…DNA was extracted from venous blood samples of each subject collected in EDTA (Ethylenediaminetetraacetic acid)-containing tubes, using a commercially available kit (Quick-gDNAMini-Prep, ZymoResearch, USA). The genotypes of the investigated polymorphism of AGT gene, namely c.-58 A>C gene polymorphism, were established by using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) technique and specific restriction enzyme as previously described (14,15,16). The inclusion criteria for both groups were: adult subjects without malignancies, with complete medical and clinical data, referred for endoscopic evaluation, with at least four routine biopsies (two from the antrum and two from the corpus) who signed the informed consent.…”
