EphB2 SNPs and Sporadic Prostate Cancer Risk in African American Men

Abstract: The EphB2 gene has been implicated as a tumor suppressor gene somatically altered in both prostate cancer (PC) and colorectal cancer. We have previously shown an association between an EphB2 germline nonsense variant and risk of familial prostate cancer among African American Men (AAM). Here we set out to test the hypothesis that common variation within the EphB2 locus is associated with increased risk of sporadic PC in AAM. We genotyped a set of 341 single nucleotide polymorphisms (SNPs) encompassing the EphB… Show more

“…For instance, the search of genes underlying the age-related macular degeneration undertaken with a small-scale GWAS using DNA samples from only 96 cases and 50 control subjects led to a prominent finding showing that a variant in complement factor H gene has an important causative effect (OR = 7.4) [46]. A case–control study of American African men on the association of EphB2 SNPs with PCa risk suggests that the power of GWAS study relies on high resolution haplotype maps based on African population [47]. Further analysis of our GWAS study should be performed when a high-resolution genetic maps of Arabs is available.…”

“…PCa patients and controls ages 40 to 79 years old were recruited from six public and academic hospitals in Chicago, IL (Cook County Health and Hospital System, Northwestern Memorial Hospital, Jesse Brown Veterans Affairs Medical Center, University of Illinois Hospital and Health Science System, and University of Chicago Hospital) between 2009 and 2014 [18] and Washington, D.C. (Howard University Hospital) between 2000 and 2004 [19]. The patients underwent their first prostate biopsy due to an elevated or abnormal serum prostate specific antigen (PSA) level or an abnormal digital rectal examination.…”

“…All participants provided written informed consent. The study and consent forms were approved by the Howard University Institutional Review Board [36, 37]. The present analysis is based on 248 men who provided demographic, lifestyle, dietary, and medical information.…”
