SummaryPorcine pleuropneumonia, caused by Actinobacillus pleuropneumoniae is one of the most important bacterial diseases of the pig respiratory tract. One hundred and forty eight Actinobacillus pleuropneumoniae strains isolated from pig's clinical samples were tested for their susceptibility to 11 antimicrobials by disk diffusion method. Isolates were obtained from the pathoanatomically altered pig's lung that died with signs of respiratory disease. Antimicrobial discs with following concentration were used: ampicillin 10 µg, amoxicillin 10 µg, amoxicillin/clavulanic acid 20/10 µg, cefotaxime 30 µg, gentamicin 10 µg, penicillin 10 IU, streptomycin 10 µg, tetracycline 30 µg, enrofloxacin 5 µg, lincospectin 15/200 µg, florfenicol 30 µg and trimethoprim/sulfamethoxazole 1.25/23.75 µg. Isolated strains showed various susceptibility to tested antimicrobials. Seventy-five percent of isolates were resistant to one or more drugs frequently used in the treatment of pig's diseases at farms in Serbia. All 148 isolates were susceptible to cefotaxime, enrofloxacin and florfenicol, while 96% of them were susceptible to amoxicillin/clavulanic acid and lincospectin. Susceptibility of isolates to amoxicillin, gentamicin and trimethoprim/sulfamethoxazole were 91%, 77% and 64%, respectively. The highest resistance rates were determined for tetracycline and streptomycin, 34% and 31%, respectively. When compared to our previous results, we noticed a decrease in resistance to tetracyclines and trimethoprim/sulfamethoxazole. Our results indicate that A. pleuropneumoniae strains isolated from pig's clinical samples in Serbia show high susceptibility to antimicrobial agents which are most often used in the pig industry.

Bacterial regulatory small RNAs (sRNAs) play important roles in gene regulation and are frequently connected to the expression of virulence factors in diverse bacteria. Only a few sRNAs have been described for Pasteurellaceae pathogens and no in-depth analysis of sRNAs has been described for Actinobacillus pleuropneumoniae, the causative agent of porcine pleuropneumonia, responsible for considerable losses in the swine industry. To search for sRNAs in A. pleuropneumoniae, we developed a strategy for the computational analysis of the bacterial genome by using four algorithms with different approaches, followed by experimental validation. The coding strand and expression of 17 out of 23 RNA candidates were confirmed by Northern blotting, RT-PCR, and RNA sequencing. Among them, two are likely riboswitches, three are housekeeping regulatory RNAs, two are the widely studied GcvB and 6S sRNAs, and 10 are putative novel trans-acting sRNAs, never before described for any bacteria. The latter group has several potential mRNA targets, many of which are involved with virulence, stress resistance, or metabolism, and connect the sRNAs in a complex gene regulatory network. The sRNAs identified are well conserved among the Pasteurellaceae that are evolutionarily closer to A. pleuropneumoniae and/or share the same host. Our results show that the combination of newly developed computational programs can be successfully utilized for the discovery of novel sRNAs and indicate an intricate system of gene regulation through sRNAs in A. pleuropneumoniae and in other Pasteurellaceae, thus providing clues for novel aspects of virulence that will be explored in further studies.

Research Article Open AccessActinobacillus pleuropneumoniae is the etiological agent of porcine pleuropneumonia, a severe, contagious pulmonary disease that causes important economic losses in industrialized pig production worldwide. A. pleuropneumoniae is a Gram-negative bacterium of the Pasteurellaceae family [1]. The importance of the infection derives from the fact that it can result in acute, chronic, or subclinical infection, causing losses due to reduced production, and increasing costs of medication and vaccination [2]. There are two biotypes of App differentiated on the basis of their requirement for nicotinamideadenine dinucleotide (NAD Actinobacillus pleuropneumoniae causes porcine pleuropneumonia, a disease which occurs world-wide and affects pigs of all ages. The study on the protective immune mechanism is not thoroughly clear. An outer membrane protein (OMP) was identified by screening a phage library of 3~8kb random DNA fragments of A. pleuropneumonia serotype 7. The inserted segment encoding the entire amino acid sequence of the OMP was determined by sequencing the rescued plasmids and BLAST analysis. Genes encoding the OMP from the isolate were amplified from genomic DNA template by PCR and cloned into a pET15b prokaryotic expression vector, generating the pET15b-OMP. Escherichia coli BL21 (DE3) competent cells were transformed with the construct followed by the induction of protein expression by the addition of IPTG. A band corresponding to the predicted size (48.766kDa) was seen on the SDS-PAGE. Polyclonal antibodies raised against recombinant OMP from Rabbits were reacted with bacterial proteins. Immunization with the recombinant OMP showed 40% mice were protected from the challenge.
