Enzyme-Structure Relationships in the Endoplasmic Reticulum of Rat Liver

Abstract: Subfractionation of preparations of rat liver microsomes with a suitable concentration of sodium deoxycholate has resulted in the isolation of a membrane fraction consisting of smooth surfaced vesicles virtually free of ribonucleoprotein particles. The membrane fraction is rich in phospholipids, and contains the microsomal NADH-cytochrome c reductase, NADH diaphorase, glucose-6-phosphatase, and ATPase in a concentrated form. The NADPH-cytochrome c reductase, a NADPH (or pyridine nucleotide unspecific) diaphora… Show more

“…Microsomes were subfractionated into ribosomes, a membrane fraction, and a microsomal content fraction following the method of Ernster et al . (18) as previously modified (8) . It is important in this procedure that the microsomes be washed thoroughly in 0 .25 M sucrose after incubation and that microsomes from at least 5 g of liver be used for each 10 ml of suspension before the addition of the 0 .26% sodium deoxycholate.…”

“…Methods have been developed, using limiting amounts of deoxycholate, which can separate the microsomes into a ribosomal fraction, a membrane fraction, and the contents of the cisternae of the microsomal vesicle (18) . In order to determine which of these microsomal subfractions contained the radio-23 6 THE JOURNAL OF CELL BIOLOGY • VOLUME 52,1972 active proteins, microsomes were incubated with either leucine-14C or the appropriate radioactive nucleotide-sugars, and then the washed microsomes were subfractionated into the three microsomal subfractions .…”

“…The rats were sacrificed at various times after the initial injection and a portion of the liver was used to make microsomes according to Ernster et al [7] . Microsomal phospholipid was extracted with chloroform-methanol [8] and lipid phosphorus was determined according 240 to Bartlett [9].…”
