Enzyme-Linked Immunosorbent Assay for the Determination of 2,4,6-Trinitrotoluene and Related Nitroaromatic Compounds

Abstract: A sensitive enzyme-linked Immunosorbent assay (ELISA) for the determination of the explosive 2,4,6-trinitrotoluene (TNT) and other nitroaromatic compounds was developed. As tracer, a trinitrophenyl-derivative conjugated to horseradish peroxidase was used. With this competitive assay, performed in polystyrene microtitre plates, TNT can be detected within the range of 0.02 µg/1 and 20 µg/ 1. Cross-reactivities at the center points (50% values) of several nitroaromatic compounds were determined. Additionally, the… Show more

“…Enzyme-linked immunosorbant assays (ELISA) and rapid immunofiltration assays for TNT in soil and water using simplified ELISAs have been described (11,12), and commercial immunoassay kits for TNT in soil (Dtech, Strategic Diagnostics) have received EPA method approval (13). These methods often involve multiple steps and analysis times ranging from 20 min to 2 h (4,(14)(15)(16)(17)(18)(19).…”

“…[30] At room temperature, the addition reaction between TNT and the SO 3 2À group can occur to form a watersoluble sulfonate, 2,4,6-trinitrobenzene sulfonic acid sodium (TNT-SO 3 Na). [31,32] According to the UV/Vis absorption spectra, the pure TNT solution was colorless (the inset image in Figure 1 a3) and had no absorption band from 300 to 700 nm (Figure 1 a3). However, when a solution of Na 2 SO 3 (33.3 mm) was presented in the pure TNT solution, the presence of a new absorption band at 436 nm clearly evidenced the formation of TNT-SO 3 Na (Figure 1 b3) and the color of the solution changed immediately from colorless to light yellow (the inset image in Figure 1 b3).…”
