Enzyme‐Linked Immunosorbent Assay (ELISA) for Detecting Antisperm Antibodies in Mice With Testicular Autoimmunity

Abstract: We developed an ELISA for measuring antisperm antibodies in the mouse by using serum samples obtained from mice immunized with murine testicular antigens in complete Freund's adjuvant (CFA) as well as from mice rendered vasectomized. Sperm antigens used were syngeneic epididymal spermatozoa and two types of soluble, murine testicular antigens prepared in our laboratory. This study deals with a) the sequential changes of antisperm antibody levels following immunization; b) determination of immunoglobulin classe… Show more

“…They suggested that there might be more limited number of these antigens in murine testes an that they are present in a highly insoluble form when compared to guinea pig antigens. Moreover, a murine testicular antigen obtained in a similar way to ASPM, only induced aspermatogenesis in about 15% of actively immunized mice (Itoh et al, 1989). Nevertheless, the consistent cellular and humoral immune response detected in rats immunized with ASPM and the high titres of specific antibodies found in rabbits injected with this immunogen (data not shown) indicate its antigenicity.…”

“…Assays of immune responses DTH to syngeneic TC or sheep erythrocytes was measured on day 40 or on day 5, respectively, by delayed footpad reaction to 1 X 10' TC or sheep erythrocytes as described [9]. Assay of anti-TC antibodies was done with the ELISA procedure at serum dilution of 1:800, and level of the serum antibodies was expressed as an absorbanee value as described [9,23].…”
