Enzyme inhibitor screening by electrospray mass spectrometry with immobilized enzyme on magnetic silica microspheres

Abstract: In this study, a novel technique for screening inhibitors by electrospray mass spectrometry (ESI-MS) with immobilized enzyme on magnetic microspheres has been demonstrated. First, the model enzyme acetylcholinesterase (AChE) is immobilized onto the 3-glycidoxypropyltrimethoxysilane (GLYMO)-modified magnetic silica microspheres. AChE activity was monitored by biochemical assay that is based on mixing of AChE immobilized microspheres and model substrate acetylcholine, separating and detecting the product through… Show more

“…The quenching solvent was found to completely stop the enzymatic reaction and be compatible with MS. NH 4 HCO 3 provided adequate buffering while being compatible with ESI and providing low ion suppression. To improve quantification, chlormequat was included in the quenching solution to act as an internal standard [28]. Typical MS spectra illustrating detection of substrate (acetylcholine), product (choline), and internal standard are shown in Figure 2.…”

“…ACh has been detected by various methods, such as liquid chromatography/electrochemical (LC-EC) method [36][37][38], liquid chromatography/electrospray ionization/ mass spectrometry (LC-ESI-MS) [8,[39][40][41][42], liquid chromatography/atmospheric pressure chemical ionization/mass spectrometry (LC-APCI-MS) [43], as well as MALDI-TOF-MS with CHCA as the matrix [12]. The performances of these methods have been listed in detail on Table 3.…”

“…In this article, a minicompound library was employed to evaluate our new methodology, which contained four known AChE inhibitors (galanthamine, tacrine, Hup A, and Hup B) [8,48] and four control chemical compounds extracted from natural herbs (ferulic acid, rutin, morin, paeonol) with no AChE inhibit effect. Each compound was tested at the molar ratio of 1:1000 (enzyme AChE:each tested compound).…”
