Development of immunoassays with broad speci®city recognition of the sulphonamide drugs has proved surprisingly dif®cult in previous work. We have taken an antiserum speci®c for sulphachlorpyridazine in an ELISA system homologous for immunogen and solid phase conjugate and used it in a heterologous ELISA format. The assay proved to have broad speci®city, recognising 15 out of 21 drugs tested. Despite low cross-reactivity (<1%) for seven of the drugs tested, a fully optimised assay could prove useful as a multiresidue method for the group of drugs. The results provide evidence regarding the binding characteristics of the antibody and the immunogenicity of the target.

The development of an enzyme-linked immunosorbent assay (ELISA) for the detection of residues of sulfachlorpyridazine (SCP) is described for the first time. The assay is highly specific for SCP, is simple to perform and has a lower detection limit of 0.65 ng/ml in assay buffer. In potential application of the assay to detect residues of SCP at the 0.1 mg/kg level in eggs, milk, beef, lamb, pork, chicken, turkey, porcine kidney, porcine liver and pig feedstuffs is discussed with regard to the effects of sample extracts on the standard curves. The antibody exhibits a rare stability in assay buffers containing up to 30% methanol. It is concluded that the ELISA for SCP has the appropriate characteristics for development into a robust method for the detection of this sulphonamide in agri-food materials.

Direct competitive enzyme-linked immunosorbent assays (ELISAs) were developed for sulfathiazole (ST) analysis. Polyclonal antibody rabbit serum adsorption to polystyrene microtiter plates was enhanced using a 1 l.tg/mL Protein A precoating solution. Detection limits (20% reduction in maximum enzyme signal) for ST in milk and honey were 12.0 and 34.7 ppb (ng/L), respectively. ST-horseradish peroxidase (HRP) conjugates prepared by linking the N4-amino group of ST to periodate-treated HRP were more efficient than those prepared using a more conventional two-step glutaraldehyde method. This represents a new method of preparing sulfonamide conjugates for use in ELlSAs.
