Both of these contain valuable procedures for enzyme analysis. Guilbault (97) has authored a chapter on fluorometric methods of analysis of enzymes, substrates, activators, and inhibitors in his book, "Fluorescence. Theory, Instrumentation and Practice." Passwater's book,(233) "Guide to the Umbelliferone (7 -hydroxy coumarin) and 4-methyl-umbelliferone are highly fluorescent compounds which have been modified to form nonfluorescent substrates for the enzymes glucuronidase, glucosidase, Ar-acetyl-/3-glucosaminidase, and ß-galactosidase. Robinson (259) used 4-methylumbelliferone^-Dglucoside as a substrate for ß-glucosidase. The substrate is split specifically by this enzyme, thus permitting the separate identification of ß-glucosidase and ß-glucuronidase in paper electrophoresis. Woolen and Turner (358, 359) assayed ß-glucuronidase in plasma using 4-methyl umbelliferylß-glucuronide as substrate, and Woolen and Walker (360), proposed 4-methyl umbelliferone-.Y-acetyl -ß-D-glucosaminide and 4-methyl umbelliferone-ß-D-galactoside as substrates for V-acetylß-D-glucosaminidase and ß-galactosidase. Likewise, Leaback (169) described conditions for the continuous fluorometric assay of Y-acetyl^-Dglucosaminidase using the same substrate, and Veritz, Gambell, and Brown (336) proposed l-naphthyl-2-acetamido-2-deox3T^-D-glucop3"ranoside as a substrate for this enzyme. Water samples of 100 ml are salted out with 35 g NaCl and extracted with 3 x 10 ml chloroform, the extract is dried and taken up with 0.2 ml ethanol. The ethanol extract is incubated with 7.857 mmol/l acetylcholinhydrochloride and a n enzymatic activity of 88.8 U/1 cholinesterase from horse-serum. An inhibition of cholinesterase and, therefore, of the formation of acetic acid from acetylcholin is caused by P-organic insecticides present in the investigated extract. Since the reaction proceeds in a buffer at pH 8.2 ... 8.3, the pH-difference occurring between sample and control constitues a measure of the cholinesterase inhibition. Prior t o reaction, an oxidation is required for thio-compounds, which is carried out with bromine water for parathion and with potassium permanganate for dimethoat. The detection limits of the method are in the nanogramrange. Extractives from plants as well as substances from faecal matter may have a disturbing effect. GrundlagenDer hohe Aufwand fur die Spurenanalytik mit Hilfe moderner Gerkte 1aBt fur bestiminte Bereiche von Forschung und Praxis rationelle Methoden zur Summenbestiiiimung nach wie vor interessant erscheinen. Fur die Untersuchung der Kontainination verschiedener Medien mit phosphororganischen Insektiziden und Carbamaten bietet sich auf Grund des toxikologischen Wirkungsmechanismus dieser Stoffe der Nachweis auf der Grundlage der Henimung der enzymatischen Aktivitat der Cholinesterase a n (ACKERMANN 1966; GIANG; VERWEIJ und BOTER; KUSSMATJL).Die Methode wurde ini Zusammenhang mit Lysimeteruntersuchungen zur Auswaschung von Insektiziden angewandt und entsprechend weiterentwickelt. Sie eignet sich in der vorliegenden Forin zur rationellen Untersuchung relativ zahlreicher Proben. J e Arbeitskraftstunde konnen etwa 3 Proben von der Extraktion bis zum Ergebnis bearbeitet werden. Die Nachweisreaktion selbst erfordert einschlieljlich der Vorbereitungen etwa 4 Minuten je Probe. Die Nachweisgrenzen liegen fur die von uns untersuchten Praparate allgeniein im Nanogrammbereich und unterschreiten damit deutlich die hygienischen Grenzwerte.
