Enzymatische Halogenierung von Tryptophan im Gramm‐Maßstab

Abstract: Beide Halogenasen chlorieren l-Tryptophan regioselektiv an der elektronisch ungünstigen C7-Position des Indolrings. Eine zusätzliche Flavin-Reduktase liefert FADH 2 , das in der aktiven Tasche der Halogenase durch molekularen Sauerstoff oxidiert wird und ein Flavinhydroperoxid bildet. Dieses wird anschließend nukleophil von einem Halogenidion angegriffen. Die dabei entstehende hypohalogenige Säure wird von der FAD-zur Tryptophanbindestelle durch einen 10 langen Tunnel geleitet.
