Triazole: A Promising Antitubercular Agent

Tuberculosis is a contagious disease with comparatively high mortality worldwide. The statistics shows that around three million people throughout the world die annually from tuberculosis and there are around eight million new cases each year, of which developing countries showed major share. Therefore, the discovery and development of effective antituberculosis drugs with novel mechanism of action have become an insistent task for infectious diseases research programs. The literature reveals that, heterocyclic moieties have drawn attention of the chemists, pharmacologists, microbiologists, and other researchers owing to its indomitable biological potential as anti-infective agents. Among heterocyclic compounds, triazole (1,2,3-triazole/1,2,4-triazole) nucleus is one of the most important and well-known heterocycles, which is a common and integral feature of a variety of natural products and medicinal agents. Triazole core is considered as a privileged structure in medicinal chemistry and is widely used as 'parental' compounds to synthesize molecules with medical benefits, especially with infection-related activities. In the present review, we have collated published reports on this versatile core to provide an insight so that its complete therapeutic potential can be utilized for the treatment of tuberculosis. This review also explores triazole as a potential targeted core moiety against tuberculosis and various research ongoing worldwide. It is hoped that this review will be helpful for new thoughts in the quest for rational designs of more active and less toxic triazole-based antituberculosis drugs.

ÖzBu çalışmada, çıkış maddesi piridin-2,5-dikarbohidrazin (2) ile tiyosemikarbazit türevleri [2,2'-(piridin-2,5diildikarbonil-bis(N-etilhidrazinkarbotiyoamit) (3B) ve 2,2'-(piridin-2,5-diildikarbonil)-bis(N-(pmetoksifenil)hidrazinkarbotiyoamit (3C)] ve bu bileşiklerin halkalaşmasından oluşan bis-1,2,4-triazol türevlerinin [5,5-piridin-2,5-diilbis(4-etil-4H-1,2,4-triazol-3-tiyol) (4B) ve 5,5'-piridin-2,5diilbis[4-(4-metoksifenil)-4H-1,2,4-triazol-3-tiyol)] (4C)] in vitro ortamda prostat kanser hücre hattı (PC-3) üzerindeki antikanser ve apoptotik etkileri ile bazı gram pozitif ve gram negatif bakteriler üzerindeki antibakteriyel etkileri belirlenmiştir. Antikanser etki sonuçlarına göre tüm maddelerin artan konsantrasyonlara bağlı olarak canlı kanser hücre sayısında azalma tespit edilmiştir. Tüm uygulanan maddelerde en yüksek sitotoksik etkinin 500 μM konsantrasyonda olduğu belirlenmiştir. Tüm maddelerin apoptozu tetiklediği belirlenmiş olup apoptotik indeks oranlarına bakıldığında en düşük apoptotik indeks oranı 4B maddesi uygulandığında, en yüksek apoptotik indeks oranı ise 3C maddesi uygulandığında tespit edilmiştir. Antibakteriyel çalışmada tüm bileşiklerin gram pozitif bakteriler üzerinde gram negatiflere nazaran daha fazla etkili olduğu görülmüştür.

Microbial diseases account for 16% of crop losses annually with fungal pests accounting for approximately 80% of that loss. Several classes of molecules have been used as fungicides for agricultural purposes. Triazole fungicides were first introduced in the mid‐1970, and they are highly effective against powdery mildews, rusts, and leaf‐spotting fungi. Currently, there are 26 commercially available triazole fungicides, with 19 having registration for agricultural use in the United States and the European Union for a variety of crops. Despite their general acceptance, triazole fungicides face regulatory hurdles due to toxicological concerns for endocrine disruption, developmental and reproductive toxicity, and liver toxicity. Further, increasing incidence of triazole resistance and changes in global weather patterns is warranting increased application rates to protect significant food crops from fungal pests. All these pressures pose challenges to farmers, agrochemical companies, and regulatory agencies which need to keep these important agricultural tools in the field while assuring safety to applicators, consumers, and the environment.
