ENU mutagenesis reveals that Notchless homolog 1 (Drosophila) affects Cdkn1a and several members of the Wnt pathway during murine pre-implantation development

Abstract: BackgroundOur interests lie in determining the genes and genetic pathways that are important for establishing and maintaining maternal-fetal interactions during pregnancy. Mutation analysis targeted to a 34 Mb domain flanked by Trp53 and Wnt3 demonstrates that this region of mouse chromosome 11 contains a large number of essential genes. Two mutant alleles (l11Jus1 and l11Jus4), which fall into the same complementation group, survive through implantation but fail prior to gastrulation.ResultsThrough a position… Show more

“…NLE plays a role in regulating the Notch activity and is involved in embryonic development in mammals by affecting the CDKN1A and Wnt pathways [ 69 ]. Forced expression of miR-26 inhibits the growth of stimulated breast cancer cells and tumor in xenograft models by reducing the mRNA expression levels of AMMECR1 and other genes [ 70 ].…”

“…cDNA was PCR amplified in triplicate and Ct values from each sample were obtained using StepOnePlus software. The relative expression of each gene was calculated based on the ΔΔCt value, where the results were normalized to the average Ct value of HPRT1 and UBC (Lossie et al, 2012).…”

“…8 Being able to isolate a factor that causes this variation (for example cell culture, PCR plate, or sex) by adding it to the statistical model increases sensitivity in detecting treatment effects and increases power. 10,11,25,30 These nuisance variables can be managed by using a randomized block design that incorporates the nuisance variables as 'blocks'. In mice, the home cage is a good example of a nuisance variable that can be used as a block to control variability from cage to cage.…”
