a b s t r a c tThis study explored the optimization of pretreatment of pineapple peel for biogas generation. Pretreatments were carried out sulfuric acid and alkaline hydrogen peroxide prior to anaerobic digestion while the response surface methodology (RSM) was used for optimization of the pretreatment procedures. The physical, chemical, proximate and structural compositions of the peels were determined prior to and at the end of the pretreatment procedures. The dynamics of microorganisms in the reactors were also evaluated by rapid molecular methods while the Fourier Transform Infra-red (FTIR) spectroscopy was employed in the identification of the chemical changes as a result of pretreatments. The use of H 2 O 2 pretreatment caused enormous lignin solubilization in the pineapple peel. In comparison, biogas production was 67% more in the alkaline pretreated pineapple peel than the biomass treated with acid and also 51% over the untreated samples. The total biogas volume produced from the acidic pretreated, alkaline pretreated, not sifted untreated and sifted untreated samples are 194.2 ± 3.0, 587.5 ± 5.2, 287.8 ± 2.1 and 245.4 ± 3.1 respectively. Thus, the alkaline pretreated experiment used lower retention time to achieve maximum gas production in this study. The use of alkaline H 2 O 2 on lignocelluloses has remained unpopular prior to this study. However, its usage in this study yielded better result than all the conventional treatments in terms of lignin solubilization and improvement in methane yield. Economically, the use of H 2 O 2 for pretreatment is adjudged feasible because the 1504 kWh t À1 TS thermal energy gain obtained from the biogas produced by the alkaline treated peel exceeded the 921 kWh t À1 TS used in the pretreatment. This gives a net thermal energy of 583 kWh t À1 TS. Whereas, the investment into acidic pretreatment of pineapple peel may not be economically justified because the total thermal energy gain of À200 kWh t À1 TS was far lower than the 1236 kWh t À1 TS thermal energy that was consumed during the pretreatment giving a net thermal energy of À1436 kWh t À1 TS. Therefore, the use of mild alkaline pretreatment is advocated in biogas generation from pineapple peel and also for biofertilizer production mostly in localities of mass production.

Microbial communities in residual slurry left after removal of stored liquid dairy manure have been presumed to increase methane emission during new storage, but these microbes have not been studied. While actual manure storage tanks are filled gradually, pilot- and farm-scale studies on methane emissions from such systems often use a batch approach. In this study, six pilot-scale outdoor storage tanks with (10% and 20%) and without residual slurry were filled (gradually or in batch) with fresh dairy manure, and methane and methanogenic and bacterial communities were studied during 120 days of storage. Regardless of filling type, increased residual slurry levels resulted in higher abundance of methanogens and bacteria after 65 days of storage. However, stronger correlation between methanogen abundance and methane flux was observed in gradually filled tanks. Despite some variations in the diversity of methanogens or bacteria with the presence of residual slurry, core phylotypes were not impacted. In all samples, the phylum Firmicutes predominated (∼57 to 70%) bacteria: >90% were members of Clostridia . Methanocorpusculum dominated (∼57 to 88%) archaeal phylotypes, while Methanosarcina gradually increased with storage time. During peak flux of methane, Methanosarcina was the major player in methane production. The results suggest that increased levels of residual slurry have little impact on the dominant methanogenic or bacterial phylotypes, but large population sizes of these organisms may result in increased methane flux during the initial phases of storage. IMPORTANCE Methane is the major greenhouse gas emitted from stored liquid dairy manure. Residual slurry left after removal of stored manure from tanks has been implicated in increasing methane emissions in new storages, and well-adapted microbial communities in it are the drivers of the increase. Linking methane flux to the abundance, diversity, and activity of microbial communities in stored slurries with different levels of residual slurry can help to improve the mitigation strategy. Mesoscale and lab-scale studies conducted so far on methane flux from manure storage systems used batch-filled tanks, while the actual condition in many farms involves gradual filling. Hence, this study provides important information toward determining levels of residual slurry that result in significant reduction of well-adapted microbial communities prior to storage, thereby reducing methane emissions from manure storage tanks filled under farm conditions.

This study evaluated the effect of the substrate:inoculum ratio and digestate recirculation conditions on the biogas and methane yield, free ammonia concentration and solid phase agronomic quality (after treatment) of poultry litter solid state anaerobic digestion (SSAD). Experimental tests were conducted using a central composite design (2²), with four trials at the factorial points and three at the central points, using poultry litter as substrate that was collected after 12 cycles of broiler production [TS 76% (w.w −1 )]. Three laboratory-scale reactors containing 3 kg of poultry litter each were operated at mesophilic conditions (37 • C) and seven experimental runs were performed at retention time of 30 days each. There were three substrate:inoculum ratios (1:1; 1:1.66, and 1:3) and three daily recirculation intervals (2, 3, and 4 times per day), and each recirculation event lasted 15 min. The highest biogas and methane yields were183 L Nbiogas .kg VSadd , respectively, and they were obtained at the substrate:inoculum ratio of 1:3 with a digestate recirculation frequency of four times a day. Digestate recirculation was the variable that influenced the concentration of free ammonia; the most frequently recirculated tests had lower concentrations (below 60 mgNH 3 .L −1 ). After the SSAD process, we identified a transfer of nutrients (N, P, K) from the solid phase to the liquid phase. The nutrient concentrations in the solid phase (N out = 11 g.kg −1 , K out = 5 g.kg −1 , and P out = 7 g.kg −1 ) indicated that the digested poultry litter still has potential for use in fertilization. In this study, it was estimated that 90 m 3 biogas .tons −1 poultrylitter was generated using SSAD. Whereas 100 tons of poultry litter are produced after 12 production cycles, the biogas generation capacity reaches 9,000 m 3 /year. In this scenario, farmers can use biogas from poultry litter SSAD as an additional source of energy.
