Enrichment and acclimation of an anaerobic mesophilic microorganism’s inoculum for standardization of BMP assays

Abstract: Appropriate enrichment of anaerobic microorganism's consortium is crucial for accurate biochemical methane potential (BMP) assays. An alternative method to produce and maintain a mesophilic methanogenic inoculum was demonstrated. Three sources of inoculum were mixed and acclimated for 857days in order to reach steady conditions (pH=7.90±0.46; VS/TS>50%; VFA/alkalinity=0.16±0.04gAcetic Acid/ [Formula: see text] ). Biogas yield >80% was obtained after 70days of inoculum acclimation in comparison to standard cell… Show more

“…This test was carried out on the pineapple peel samples so as to determine the potential biogas yield from the substrate under standard condition. The equipment for this test was 250 mL batch mini-reactors connected to 500 mL eudiometer tubes with 10% (m.v À1 ) VS at 37 C for 30 days [47] along with the VDI 4630 (2006) standard. Increment and stabilization of the biogas yield was achieved when biogas production per day was lower or equal to 1% of the total production from the entire process.…”

“…With long-term storage of dairy manure, bacterial and methanogenic communities of different sources (e.g., rumen, soil, water, and bedding material) may be enriched and acclimatized to storage conditions (9,10). All microbial communities functioning in rumen environments may not be able to establish in manure storage systems, as these environments can differ with a number of parameters, such as temperature and substrate composition.…”

“…The inoculum used for SSAD was collected from a standardized mesophilic acclimated inoculum from a lab-scale CSTR (continuous stirred-tank reactor), according to the methodology described by Steinmetz et al (2016). The acclimated inoculum was a mixture of equal volumes of anaerobic sludge from two independent large-scale UASB (Upflow Anaerobic Sludge Blanket) reactors (swine wastewater; gelatin manufactory effluent) and from fresh dairy cattle manure.…”
