Enhancing the Anticancer Activity and Selectivity of Goniothalamin Using pH-Sensitive Acetalated Dextran (Ac-Dex) Nanoparticles: A Promising Platform for Delivery of Natural Compounds

Abstract: Goniothalamin (GTN), a natural compound isolated from Goniothalamus species, has previously demonstrated cytotoxic activity against several cancer cell lines. However, similarly to many natural and synthetic anticancer compounds, GTN presents toxicity toward some healthy cells and low aqueous solubility, decreasing its bioavailability and precluding its application as an antineoplastic drug. In our efforts to improve the pharmacokinetic behavior and selectivity of GTN against cancer cells, we developed a polym… Show more

“…For cytotoxicity assessment, WI-38 normal human fibroblast cells were used for the calculation of SI which is the ratio of IC 50 retinoid (WI-38)/IC 50 retinoid (cancer cell line) [ 134 ]. Increasing SI value above 1 indicates a more effective and safer drug as an anticancer compared to normal tissues [ 135 ].…”

“…Although the basis of EPR effect is not completely understood and still in dispute, [97][98][99][100] the Ac-DEX-based delivery systems have shown some promising results in vivo. 101,102 Below, we briefly introduce the application of Ac-DEX-based delivery systems in different cancer therapies, and highlight some novel designs.…”

“…It is also interesting to consider structural changes in the microtubule network induced by invading viruses [30] as well as the topology of microtubular network [31]. Our model opens new avenues for the quantitative analysis of the endosomal escape of a virus-mimicking pH-responsive nanoparticle for anti-tumor therapeutics delivery [32][33][34][35].…”
