Short-day treatment has been used for some conifer species to improve planting stock quality. The aim of this paper was to study the effect of short-day treatment on performance and quality of young seedlings of Italian cypress (Cupressus sempervirens L.), a conifer species abundant in the Mediterranean area. Seeds of Italian cypress were sown in mini-plug cavities. After 5 weeks of growth, seedlings were subjected to four different photoperiods: (a) 14 h (control), (b) 8 h, (c) 6 h, and (d) 4 h for a period of 1-3 weeks, before they were transplanted to bigger size containers. Reducing photoperiod did not result in bud formation or shoot inhibition although it increased the number of lateral shoots. Seedlings subjected to 6-h photoperiod for 2 weeks during pre-cultivation in mini-plugs had improved root characteristics, giving them a higher potential for outplanting success, especially in Mediterranean areas.

For successful afforestation programs seed quality is crucial, but seedlings are susceptible to climatic stress. Therefore, to improve afforestation success it is necessary to compare performance of seedlings from natural and cultivated populations under different climatic conditions. We investigated growth performance in seedlings of three natural and four afforested Syrian Cupressus sempervirens L. populations under different temperature and moisture regimes. A "warm" climate chamber approximately simulated current mean annual temperatures (day/night: 20/10°C) while a "hot" chamber simulated an average increase of 5°C (day/night: 25/15°C). Seedlings were irrigated twice (drier) or thrice (moist) weekly. Seedlings from natural provenances outperformed those from afforested stands in all growth variables in both chambers. In the warm chamber, root length and biomass were not affected by irrigation for both population types, but shoot height decreased for afforested seedlings under drier treatment while it slightly increased in natural seedlings. In the hot chamber, shoot height decreased but root length and biomass increased for population types under the drier treatment. Comparison between the two chambers showed that under the drier treatment shoot height and biomass decreased at higher temperatures, but root length and biomass were not significantly different. The same response to higher temperatures was observed under the moist treatment, but root biomass decreased too. Our results emphasize the necessity to protect the remaining natural forest of C. sempervirens in Syria and recommend systematic collection of seed material from natural stands for afforestation programs. This might also hold for ex situ cultivation of retrieving rare and endangered plant species.

Trisetacus juniperinus (Nalepa) sensu Keifer (Acari: Eriophyoidea: Phytoptidae) causes irregular development of buds, shoot deformations and stunted growth of trees, resulting in a serious threat to nurseries and young stands of Cupressus sempervirens L. (Mediterranean cypress). Recently, some cypress clones selected for their resistance to the fungal canker agent Seiridium cardinale (Wag.) have shown high susceptibility to the mite. Considering its tiny body, its hidden lifestyle inside the buds and the probable occurrence of other species (the vagrant Epitrimerus cupressi (Keifer) is common on the Mediterranean cypress in Italy), detection and monitoring of T. juniperinus require taxonomic expertise and are often time-consuming and challenging before serious damage is discernible. In the present study, a rapid, cost-effective PCR-based method was developed and validated to detect T. juniperinus on cypresses. The cytochrome c oxidase subunit I gene was amplified with degenerate and specific primers, but the latter were the only ones able to discriminate between T. juniperinus and E. cupressi. PCR products distinguished the two species both in a pool of individuals in a mixed population of both species and in single individuals, indicating the sensitivity of the detection method. PCR-RFLP (restriction fragment length polymorphism) by means of XmnI and XbaI endonucleases separated the two species. Furthermore, a washing-sieving protocol was used to make mite collection from the tree sample faster and simpler; this procedure did not interfere with the molecular detection of the species. The possibility of the routine use of this assay to monitor quarantine eriophyoids infesting plant material is discussed.
