Enhancing Efficacy of Stem Cell Transplantation to the Heart with a PEGylated Fibrin Biomatrix

Abstract: Bone marrow-derived mononuclear cell (BMNC) transplantation provides the possibility of rescue or regeneration of myocardium lost during acute myocardial infarction (AMI). The extensive death of transplanted cells and the lack of sustained engraftment may limit its application. We investigated whether delivery of BMNCs by an injectable PEGylated fibrin biomatrix that covalently binds hepatocyte growth factor (HGF) would enhance the rate of cell engraftment and improve cardiac function. Balb/C female mice with … Show more

“…[20][21][22] Human fibrinogen (F3879; SigmaAldrich, St Louis, MO) was solubilized in phosphate-buffered saline (PBS, 21-030-CV; Corning, Manassas, VA; adjusted to pH 7.8) at a concentration of 80 mg/mL. After 2 h of incubation at 37°C and brief vortexing, the solution was sterilized using a 0.20-mm SteriFlip filter (EMD Millipore, Billerica, MA).…”

“…[2][3][4][5] Previous work by our group demonstrated that PEGylated fibrin gels promote mesenchymal stem cell (MSC) tubulogenesis and differentiation towards a vascular cell type. 4 These findings suggest MSCs could potentially be d elivered to an injury site after an ischemic event in order to assist in neovascularization and tissue repair.…”

“…After administration in a mouse MI model, the systems allowed significant improvements in cell prevalence at the injection site for at least 4 weeks, compared to free cell administration. Interestingly, in BMSC-HGF-hydrogel treated animals, cell retention was accompanied by the lowest levels of apoptosis and the highest LV function recovery among all the groups, confirming that tissue engineering was more effective than protein or cell therapy alone [150].…”
