Enhancing Application Efficiency of Pseudomonas SPP. and Serratia marcescens Isolates against Meloidogyne incognita in Tomato Plants.

A survey of nematode community in grapevine plantations grown in Belbes, Sharqia, Egypt revealed the presence of nine plant-parasitic nematodes genera namely Aphelenchus, Criconemoides, Ditylenchus, Helicotylenchus, Pratylenchus, Meloidogyne, Longidorus, Tylenchus and Tylenchorhynchus; free-living nematodes (FLNs) belonging to the genus Rhabditis and predacious nematodes (PNs) belonging to order Mononchida. Nematodes were found with a different frequency of occurrence and population density. Pratylenchus gained a high population density (PD) followed by Tylenchus, Meloidogyne and Helicotylenchus. A higher prominence value (PV) was found with genus Pratylenchus (190.75) while, the genera Meloidogyne, Tylenchorhynchus and Tylenchus were found with moderate prominence values. Animal manures are effective tools in sustainable soil health and agricultural systems. Therefore, the influence of three livestock manures as feasible practices with BECTO Grow Roots®, a commercial plant growth-promoting bacterium (PGPB) on nematode community and plant growth and fruit yield of grapes, Vitis vinifera L. cv. Flame was undertaken. Soil samples were collected at five-time intervals. Composted animal manures significantly (P ≤ 0.05) minified numbers of plant-parasitic nematodes (PPNs). The greatest effect was recorded with oxamyl after two months of treatments followed by chicken, sheep and cow manure. Conversely, at four and five months after application, animal manures were the most effective. Moreover, a noticeable abundance of FLNs and PNs numbers was detected. The highest increment was achieved in soil amended with chicken manure followed by sheep and cow manure whereas oxamyl reduced numbers of FLNs and PNs as compared with untreated vines. Application of manures against PPNs particularly M.incognita decreased numbers of J 2 /250/soil in vines amended with chicken, cow and sheep manure, respectively and increased numbers of FLNs and PNs. Population densities of M.incognita J 2 /250 g soil were significantly minimized (1.55 J 2 /250 g soil) in blocks amended with commercial rhizobacteria and chicken manure. Maximum total fruit weight and leaf numbers were achieved in vines treated with rhizobacteria +chicken, cow and sheep manure, respectively. This study proved that composted chicken, cow and sheep manure applied alone or in combination with PGPB can be a promising tool for the management of phytoparasitic nematodes, enrichment of FLNs and PNs and improvement of plant growth and grapevine yield.

Effect of Different Control Agents on Meloidogyne Incognita Kofoid Infesting Cucumber Plants

The stock solution 10% and half stock solution 5% of three oils namely; mineral oil (Diver oil 97 % EC), plant oils marjoram oil (Origanum majorana L.) and orange oil (Citrus aurantium L.), as well as two species of bacteria Bacillus megaterium and BTS1 (Bacillus poylmyxa) at two concentrations showed nematicidal activity against egg-mass and second stage juveniles of Meloidogyne incognita (Kofoid and White) Chitwood in vitro experiments. Egg hatching and juvenile mortality were significantly (P ≤ 0.05) influenced by tested materials, concentration and exposure time. B. megaterium , diver oil and marjoram oil gave a higher effect, while B. poylmyxa was the lowest effective one. In case of juvenile mortality, B. poylmyxa , diver oil and marjoram oil gave the highest percentage of juvenile mortality (75.14, 73.00 and 61.85 %) after exposure to stock solutions (10%) and (53.00, 67.42 and 52.57 %) when exposed to half stock solution (5%) after seven days of treatment, respectively. Under greenhouse conditions, results indicated that, oxamyl, dry leaf powder of moringa (Moringa oleifera) and B. megaterium were the most effective in suppressing root galling of M. incognita infecting cucumber plants. Minimum number and gall size particularly ≥ 4mm was recorded in cucumber plants treated by oxamyl, moringa and B. megaterium. Data showed that moringa and B. megaterium could be used to increase crop yield of cucumber plants and for controlling rootknot nematode, M. incognita.

Background Acceptable alternative eco-friendly tools in the present study were tested to control the root-knot nematode, Meloidogyne incognita, on greenhouse-cultivated vegetables. The nematicidal effect of rhizobacteria (Pseudomonas and Serratia), egg parasitic fungus (Purpureocillium lilacinum), abamectin (Streptomyces avermitilis), and 3 botanicals (colocynth, Citrullus colocynthis; moringa, Moringa oleifera; marigold, Tagetes erecta L.) singly or in combination was tested against M. incognita, in comparison with emamectin benzoate. Results In vitro treatments revealed that egg hatching and juvenile mortality were influenced by the type of bioagents, plant species of botanicals, and exposure time. All the tested bioagents and botanicals displayed nematicidal potential via their ovicidal and larvicidal action on egg hatching and J2 mortality of M. incognita. Three and 5 days post-treatment, abamectin and emamectin benzoate were more effective than P. lilacinum, Serratia and Pseudomonas, and C. colocynthis in inhibiting egg hatching: 96.31 and 94.88%; 95.79 and 94.05%; 94.11 and 94.46%; 85.54 and 87.28%; 88.87 and 84.30%, respectively. On the other hand, after 10 days, P. lilacinum gave the highest inhibition percentage (99.00%), followed by abamectin (89.25%). However, the difference was insignificant compared with the inhibition percentage of rhizobacteria, Serratia and Pseudomonas (88.69%; p ≤ 0.05). Moreover, juvenile mortality was 100.0, 96.80, and 91.60% after 10 days of treatment, respectively. However, botanicals showed a lower effect on egg hatching and juvenile mortality. Under greenhouse conditions, potential antagonism towards M. incognita by application the mixture of biocontrol agents and botanicals was more effective in controlling M. incognita than single treatments. Conclusions The combination of abamectin and/or emamectin benzoate with P. lilacinum and rhizobacteria was the most effective against M. incognita, followed by rhizobacteria and P. lilacinum, not only in decreasing galls and reproduction of M. incognita but also in increasing plant growth of tomato parameters than the control. The application of various bioagents including abamectin might be a potential antagonism strategy against phytonematodes in protected agricultural areas.
