Lipoglycans (lipopolysaccharides, lipoarabinomannans and glycolipids) are unique components of the cell membrane of all cells and the envelope of many bacteria. They play important roles in determining cell-environment interactions, which, however, are only partly understood due to incomplete description of their structural components, lipids and glycans. Capillary electrophoresis is an analytical technique of high separation efficiency and minimum sample requirements and has successfully been used for the analysis of several molecules of biological importance: proteins, nucleic acids and glycoconjugates. In the last years, a few applications of capillary electrophoresis to the analysis of lipoglycans have been reported. Analysis of lipoglycans involves the study of two parameters: intact molecules and carbohydrate parts. The conjunction of capillary electrophoresis and mass spectroscopy not only enhances the detection sensitivity, but also provides structural information on these structurally complex molecules. The interest in the field is rising and the results from the exact determination on the lipoglycan structure are expected to improve our understanding of the molecular mechanism of lipoglycan binding to proteins and cells of host organisms as well as their relationship to the virulence and pathogenesis of bacteria. In this report, an overview of the capillary electrophoresis methods used to analyze and characterize the intact lipoglycans as well as their carbohydrate parts is presented.

Overview on advances in capillary electrophoresis-mass spectrometry of carbohydrates: A tabulated reviewThe increasing interest for carbohydrates as holder of essential bioinformations has boosted their full characterization through analytical techniques. The intent of this review is to summarize the recent trends regarding on-line and off-line CE-MS coupling for carbohydrate analysis. A statistical survey on the articles that use derivatizing agents as well as on the analyzer and type of instrument coupling (i.e. on-or off-line) is depicted. From a general overview it can be concluded that, whereas derivatization might be useful for the detection of neutral carbohydrates improving separation selectivity with volatile buffers and increasing sensitivity of the MS detection, relatively few works with derivatized carbohydrates were found; this was noticed in particular for glycosides and saccharides carrying ionizable groups, which are normally analyzed without any chemical modification. The most applied coupling is the on-line sheath-liquid interface; for online applications, ESI is the sole source used, whilst the most common analyzer is the IT. MS n is often exploited, as fragmentation increases the achieved structural information. CE-MS turned out to be mainly used for the analysis of carbohydrates in drug development (i.e. study of oligosaccharides from pathogens, carbohydrate-based drugs and drug metabolites), in nutrition and for characterization of glycans from glycoproteins. The reader will find elucidating tables regarding these recent CE-MS applications, including the main information on the analysis conditions. Comments are meant to help the immediate focus on the usefulness of the analytical technique and predict the difficulties found during analysis and, in case, their overcoming.

AbstractIn the present work, analytical methods for determination of neutral species based on capillary electrophoresis (CE) with contactless conductivity detection (CCD) were developed. Free solution capillary electrophoresis (FSCE) and micelar electrokinetic chromatography (MEKC) were applied to three cases: mixtures of aliphatic alcohols, mixtures of sugars, and salbutamol. These substances were selected due to their distinctiveness. The alcohols have well-defined polar and apoIar regions in their carbonic chain and are typical non-ionic species in aqueous medium. The sugars have a great number of hydroxyl groups along the carbonic chain, which give a polar nature and can be ionized at high pH values. The salbutamol is amphoteric. The best approach for the a1cohols was MEKC that allowed separation of five pentanol isomers with limit of detection of 10-4moI,L-I. A running electrolyte with pH 12.1 allowed determination of 10-5moI,L-I of fructose, glucose, galactose, and sucrose, using FSCE with 20-/-lminner diameter capillary. The sensitivity for the cationic and anionic forms of albuterol in FSCE was about ten times greater than that one achieved by MEKC for the neutral species. Since the conductivity detection in MEKC was not c1ear1yunderstood, some experiments were carried out in order to improve the knowledge about it. Conductivity and viscosity measurement as well as impedance analysis of buffered sodium dodecyl sulfate solution with n-alcohols and experiments using similar conditions in MEKC showed that CCD response is complex, because several phenomena contribute to the solution conductivity in the region of the analytes. The main phenomena, which are not proper1y considered in the previous models, are the effects of the analytes on the dissociation and ionization of the micelles. These phenomena would be responsible for the vacancies detected as negative peaks, which would be formed by the augment of the mobilities of the species involved in the micelle formation. lU
