Enhancement of hyperthermia-induced apoptosis by 5Z-7-oxozeaenol, a TAK1 inhibitor, in A549 cells

Abstract: KRAS mutant lung cancers have long been considered as untreatable with drugs. Transforming growth factor-β-activated kinase 1 (TAK1) appears to play an anti-apoptotic role in response to multiple stresses and has been reported to be a responsive kinase that regulates cell survival in KRASdependent cells. In this study, in order to find a useful approach to treat KRAS mutant lung cancer, we focused on the combined effects of 5Z-7-oxozeaenol, a TAK1 inhibitor, with hyperthermia (HT) in KRAS mutant lung cancer ce… Show more

“…Research on the effect of TAK1 inhibitors on apoptosis has also been a hot topic in recent years. It has been reported that a TAK1 inhibitor promotes the apoptosis of cervical, ovarian and lung cancers, as well as lymphoma cells [21,30,31]. As a common protein kinase in multiple signal transduction pathways in cells from different tissues, TAK1 or its inhibitor may activate different signalling pathways, and this will be the focus of our follow-up study.…”

“…They transduce the apoptotic cell death signals in a cascade manner, where the initiator caspases (caspase-8) cleave and activate the effectors (caspase-3), which then leads to cell apoptosis. 18 , 35 , 36 As zymogen caspase-3 exists in the cytoplasm and is activated in the early stage of apoptosis to degrade the substrates in cells, eventually leading to apoptosis. 37 …”

“…As expected, Ep-ME significantly inhibited the phosphorylation of TAK1 and dramatically downregulated the mRNA expression of MMP1, MMP2, and MMP9 in HEK293T cells overexpressing TAK1 (Figure 4a,b). Subsequently, we tested the effect of the resorcyclic acid lactone TAK1 inhibitor 5Z-7-oxozeaenol on inflammation [22]. The 5Z-7-Oxozeaenol decreased the NO production of LPS-treated RAW264.7 cells in a dose-dependent manner and had no significant cytotoxicity in the concentration range from 20 nM to 80 nM (Figure 4c,d).…”
