Background Despite the well-known effect of type-I collagen in promoting cartilage repair, the mechanism still remains unknown. In this study we investigated this mechanism using a rabbit model of cartilage defects.Animals and methods 5-mm-diameter full-thickness defects were created on both patellar grooves of 53 Japanese white rabbits (approximately 13 weeks old). The left defect was filled with collagen gel and the right defect was left empty. The rabbits were killed and examined morphometrically until the twenty-fourth postoperative week, by (1) evaluation of matrix production, (2) enumeration of the total number of cells engaged in cartilage repair, (3) enumeration of the proliferating cells, (4) localization of mesenchymal stem cells, and (v) localization of apoptotic cells.Results We found that type-I collagen enhances cell recruitment, and thereby increases the number of proliferating cells. A considerable proportion of the proliferating cells were identified as bone marrow-derived mesenchymal stem cells. However, type-I collagen does not prevent the chondrocyte precursors from undergoing apoptotic disengagement from the chondrogenic lineage.Interpretation Type-I collagen promotes cartilage repair by enhancing recruitment of bone marrowderived mesenchymal stem cells. Additional use of agent(s) that sustain mesenchymal stem cells along the chondrogenic path of differentiation may constitute an appropriate environment for cartilage repair.

We described in-vivo potential of collagen type I scaffold extracted fromknee of ostrich. Full thickness defects were created bilaterally in the weight bearing area of femoral condyles of the both femurs of 7 white New zealand adult male rabbits weighting 1800±100 g. The six left medial condyles as experimental group I were filled by using cell-free tissue and six right medial condyles of the same rabbit were treated by cell-tissue as experimental group II and the right medial condyle defect of one rabbit as control group was left without treatment. Histopathology data demonstrated significant difference between three groups with respect to the cartilage repair indicators as the morphology of the cells, color matrix and cartilage thickness (P<0.05). Furthermore, sub-chondral bone formation showed a significant difference and relationship between two experimental groups and control group (P<0.05). The use of cell-free ligamentum plantar matrix showed markedly significant improvement in the processes of joint cartilage repair compared control group (P<0.05). Results support the use of cell-free ligamentum plantar can be useful and promising bio-engineering procedure as a scaffold for the treatment of the articular cartilage surface. Bu çalışmada devekuşunun dizinden ekstrakte edilen kollajen tip I doku iskelesinin in vivo potansiyeli tanımlanmıştır. Ağırlıkları 1800±100 g olan 7 adet Yeni Zelanda ergin erkek tavşanın her iki femurunda femoral kondulusların ağırlık taşıyan bölgelerinde bilateral olarak tüm kat hasarı oluşturuldu. Altı sol medial kondulus hücre içermeyen doku ile doldurulurken (Grup I) aynı tavşanların altı sağ medial kondulus hasarı hücre içeren doku ile tedavi edildi (Grup II) ve bir tavşanın sağ medial kondulus hasarı tedavi uygulanmayarak kontrol grubu olarak bırakıldı. Histopatoloji bulguları kartilaj onarım belirteçleri olarak hücre morfolojisi, renk matriks ve kartilaj kalınlığı bakımlarından her üç grup arasında anlamlı derecede fark olduğunu gösterdi (P<0.05). The purpose of this study was to evaluate the (ultra-) morphology, biochemical behavior, and activity of ovine chondrocytes seeded on a matrix consisting of three types of collagen (I, II, III) with two different degrees of UV crosslinking. Ovine articular chondrocytes were isolated from stifle joints and seeded on both types of UV-crosslinked collagen matrices, as well as on non-crosslinked sponges, and cultured for 12 h, 7 days, 14 days, and 21 days. Histological analysis, electron microscopy, biochemical assays for glycosaminoglycans, and real-time quantitative PCR for collagens were performed for cell-seeded and unseeded matrices. There was no dramatic difference in the morphology and bioactivity of the cells; however, concerning handling characteristics and integrative stability, both types of crosslinked sponges were superior to non-crosslinked constructs. The results demonstrate that ovine articular chondrocytes express their phenotype in a sponge consisting of collagen; furthermore, because of mechanical reasons, non-crosslinked matrices cannot be recommended for implantation in vivo.
