Enhanced selective cellular proliferation by multi‐biofunctionalization of medical implant surfaces with heterodimeric BMP‐2/6, fibronectin, and FGF‐2

Abstract: Increasing cell adhesion on implant surfaces is an issue of high biomedical importance. Early colonization with endogenous cells reduces the risk of bacterial contamination and enhances the integration of an implant into the diverse cellular tissues surrounding it. In vivo integration of implants is controlled by a complex spatial and temporal interplay of cytokines and adhesive molecules. The concept of a multibiofunctionalized TiO 2 surface for stimulating bone and soft tissue growth is presented here. All s… Show more

“…S11D). Particularly, bFGF is known to induce proliferation of fibroblasts and SMCs [ 20 , 36 ] and to interplay with the cytokine IL1B in many infectious and inflammatory diseases [ 31 , 59 ]. EV secretion processes represent cell–cell communication and are also important for cytokine exchange [ 21 ].…”

“…It was shown by different groups that cell behavior is directly connected to their environment, meaning that culturing cells in 2D or 3D substrates makes an enormous difference [287]. Two-dimensional substrates, such as the classical polystyrene culture dishes, are, from a mechanobiological point of view, relatively simple, as they are easily characterized by stiffness (Young's modulus), the material's physiochemical properties (composition), and ligand density in the case of biofunctionalized materials [288,289]. Contrary to that, 3D materials offer adhesion points in all three dimensions and further show differences in porosity, microarchitecture, and local rigidity.…”

“…Ettelt et al used a biotin-streptavidin multilayer system to immobilize BMP-2/6 heterodimer with fibronectin. The system prevents the side effect from growth factor over-dosage and demonstrates enhanced selective osteoblast proliferation [94].…”
