Enhanced Proteolysis of β-Amyloid in APP Transgenic Mice Prevents Plaque Formation, Secondary Pathology, and Premature Death

Abstract: Converging evidence suggests that the accumulation of cerebral amyloid beta-protein (Abeta) in Alzheimer's disease (AD) reflects an imbalance between the production and degradation of this self-aggregating peptide. Upregulation of proteases that degrade Abeta thus represents a novel therapeutic approach to lowering steady-state Abeta levels, but the consequences of sustained upregulation in vivo have not been studied. Here we show that transgenic overexpression of insulin-degrading enzyme (IDE) or neprilysin (… Show more

“…197,198 An age-related reduction in neprilysin expression has also been reported. 199 Recently, a number of studies have suggested that environmental enrichment and physical activity may reduce Ab load and lower the risk of developing AD in animal models 200,201 and epidemiological studies, 202,203 respectively.…”

“…Overexpression of NEP or IDE in neurons of transgenic mice showed reduction in the levels of soluble Ab, prevention of amyloid plaque formation, and extension of lifespan (Leissring et al, 2003). We determined whether social isolation affected NEP and IDE by immunoblotting the expression of IDE and NEP with anti-IDE and anti-NEP antibodies in the APP/PS1 mice.…”

“…These findings might be clinically relevant given the fact that neprilysin expression in the brain decreases during aging and in the early stages of AD progression (Yasojima et al, 2001;Iwata et al, 2002), whereas elevated neprilysin activity reduces the accumulation of both soluble and fibrillar A in APP transgenic mice (Leissring et al, 2003;Iwata et al, 2004).…”
