Enhanced productivity of protease-sensitive heterologous proteins by disruption of multiple protease genes in the fission yeast Schizosaccharomyces pombe

Abstract: The creation of protease-deficient mutants to avoid product degradation is one of the current strategies employed to improve productivity and secretion efficiency of heterologous protein expression. We previously constructed a set of single protease-deficient mutants of the fission yeast Schizosaccharomyces pombe by respective disruption of 52 protease genes, and we succeeded in confirming useful disruptants (Idiris et al., Yeast 23:83-99, 2006). In the present study, we attempted multiple deletions of 13 prot… Show more

“…The S. pombe strain ARC010-1 (h − leu1-32 ura4-D18) (Idiris et al 2006) was used for this study. S. pombe strains were grown in EMMS or EMMS dropout media depending on the selective pressure required to maintain the plasmids (Alfa et al 1993).…”

“…S. pombe is particularly attractive, having served as a model organism for the development of metabolic engineering strategies to produce foreign gene products and certain metabolites (Giga-Hama et al 1994;Isoai et al 2002;Ikeda et al 2004;Giga-Hama et al 2007;Takegawa et al 2009;Idiris et al 2006Idiris et al , 2010a. Also, its fatty acid composition is simple.…”

“…One promising approach to overcome this difficulty is the creation of appropriate protease-deficient mutants. For example, the production of protease-sensitive human growth hormone or human transferrin was found to be significantly enhanced in S. pombe strains that lack single or multiple proteases [46][47][48][49]. In addition, it has been previously reported that modification of the growth medium conditions clearly improved the secretion efficiency [37,50].…”

“…In the fission yeast Schizosaccharomyces pombe , disruption of multiple protease genes, including ppp80 / atg4 , enhances the productivity of human growth hormone (hGH) [35]. However, the atg4 gene was one of seven protease genes that were disrupted based on a search for such genes in the S. pombe genomic database, but the functional effects of atg4 disruption on hGH production were not examined.…”
