Enhanced Ocular Anti-Inflammatory Activity of Ibuprofen Carried by an Eudragit RS100® Nanoparticle Suspension

Abstract: Purpose: To investigate the ocular pharmacodynamic profile of a polymer nanoparticle system loaded with sodium ibuprofen (IBU-RS) in comparison to an aqueous solution of ibuprofen lysinate (IBL) in the rabbit eye both being applied topically. Methods: Ocular inflammation was elicited by topical application of sodium arachidonate. Inflammation was quantified according to a modified Draize test. The protein level and the number of polymorphonuclear leukocytes in the aqueous humor were assessed after 2 h from ara… Show more

“…It was suggested that the components forming the nanocapsules act as penetration enhancers or as an endocytotic stimulator. The investigation of the ocular pharmacodynamic profile of ibuprofen-loaded Eudragit nanoparticles also leads to the conclusion that the nanosystem increased the bioavailability of the anti-inflammatory drug and ultimately its pharmacological activity (359).…”

“…62 Some studies discussed immune system stimulation by methacrylate nanoparticles. 20,[62][63][64][65][66] Although IL-12 stimulation by nanostructures without peptide has been observed (Figure 4), this same stimulus was not observed when nanostructures were associated with clavanin, probably because clavanin A suppresses the effect as shown in Figure 4. Furthermore, it is important to report that besides IL-12 stimulation by nanostructures without peptide, no cytotoxic effects against erythrocytes were observed.…”

“…71 In an in vivo study using rabbit eye, EUDRAGIT nanosuspension containing 0.1% ibuprofen showed a very good ocular tolerability. 33,72 It is useful to understand anatomical differences between species for interpreting toxicological and pathological responses to the eye and for human risk assessment of these new therapies.…”
