Enhanced Glucocorticoid Feedback Inhibition of Hypothalamo-Pituitary-Adrenal Responses to Stress in Adult Rats Neonatally Treated with Dexamethasone

Abstract: We studied the long-term effect of neonatal treatment with the synthetic glucocorticoid dexamethasone (DEX) on stress responsivity later in life. It was found that the plasma adrenocorticotropin hormone (ACTH) and corticosterone (CORT) responses induced by novelty or conditioned fear stress were markedly attenuated in adult rats that had been neonatally treated with DEX as compared with saline (SAL)-treated controls. Since there were no differences in the heart rate, body temperature, plasma noradrenaline, pla… Show more

“…with Dex (Dexamethasone Sodium Phosphate; BUFA, Uitgeest, The Netherlands) on d 1 (0.5 mg/kg body weight), d 2 (0.3 mg/kg body weight), and d 3 (0.1 mg/kg body weight) after birth. This treatment was based on a 21-d tapering treatment given to preterm infants to prevent or reduce CLD (19). Rat pups treated with equal volumes (10 L/g body weight) of sterile pyrogen-free saline (Sal) at d 1, 2, and 3 served as control group.…”

“…In 2000, two papers were published that cautioned the use of Dex during the neonatal period in humans (10,11). Since then, more adverse effects of neonatal GC have been shown in rodents on the immune (12) and neuroendocrine systems (13), on the NMDA receptor (14) and in human follow-up studies of prematurely born children treated with Dex during the neonatal period (15,16).…”

“…While there is evidence that perinatal exposure to high dose glucocorticoids may permanently re-programme HPA axis function (18)(19)(20), the induction of prolonged or permanent changes in HPA axis function by glucocorticoid exposure in adulthood is more open to speculation. Most studies have used normalization of individual secretory components of the HPA axis following exogenous stimulation to signal complete recovery after glucocorticoid withdrawal and have not considered the underlying role of the mitotic and apoptotic elements of the response.…”
