In the present manuscript, we report on the proteolytic enzymes acting in the Araucaria bidwillii megagametophyte throughout seed germination. At seed maturity the megagametophyte contains a bulk of reserves for the growing embryo, thus representing the major storage tissue of the bunya pine seed. Soon after seed germination the megagametophyte undergoes storage protein mobilization, degenerating as a no longer needed tissue by the late germinative stages. By using in-solution and in-gel assays, and mass spectrometric analyses we detected exopeptidases and proteinases differently active in this tissue at selected germinative stages, and obtained preliminary data on the nature of an endopeptidase active at the late stages. Early germination stages were characterized by aminopeptidase and aspartic, metallo and cysteine proteinase activities; carboxypeptidases and serine proteinases became highly active by the late stages. Partial sequencing of a protein responsible for late stage serine peptidase activity sensitive to the caspase-6 inhibitor, showed a set of amino acid sequences with various degrees of identity with various plant subtilisin-like serine proteinases. The participation of the early stage proteases in the storage protein mobilization and the involvement of the late stage proteases in the megagametophyte cell death are proposed and discussed.

Aedes aegypti transmits the viruses that cause yellow and dengue fevers. Vector control is essential, since a vaccine for dengue has not as yet been made available. This work reports on the larvicidal activity of Myracrodruon urundeuva leaf lectin (MuLL) against A. aegypti fourth-stage larvae (L(4)). Also, the resistance of MuLL to digestion by L(4) gut proteases and the effects of MuLL on protease, trypsin-like and α-amylase activities from L(4) gut were evaluated to determine if lectin remains active in A. aegypti gut and if insect enzyme activities can be modulated by MuLL. MuLL promoted mortality of L(4) with LC(50) of 0.202 mg/ml. Haemagglutinating activity of MuLL was detected even after incubation for 96 h with L(4) gut preparation containing protease activity. MuLL affected the activity of gut enzymes, inhibiting protease and trypsin activities and stimulating α-amylase activity. The results suggest that MuLL may become a new biodegradable larvicidal agent for dengue control. Larvicidal activity of MuLL may be linked to its resistance to proteolysis by larval enzymes and interference in the activity of digestive larval enzymes.

Forcing is a method that is usually used to induce flowering in plants, independent of the natural blooming period. The temperatures required to start blooming in lilac in November are ca. 37°C causing degeneration of flowers. Forcing at 15°C in November requires 49 days to bloom as compared to 23 days for the standard 37°C, but gives panicles of much better quality than those forced by standard method (37°C). In this study, we have investigated the protein content, total protease activity, and cysteine protease activity at different stages of flowering (flower bud whitening and swelling, open flowers, wilted flowers) for lilac flowers (Syringa vulgaris L., fam. Oleaceae) blooming under three different conditions: natural conditions in May and forcing in a greenhouse in November at 37°C (standard forcing) or at 15°C (alternative forcing). The protein content was relatively constant during flowering for each of the three sets of conditions. Flowers from 15°C had a significantly lower total protease and cysteine endoprotease activity than flowers from 37°C at all stages. Flowers from plants blooming in May had a very time-specific cysteine protease activity, which was dramatically higher for the open flower stage than for the other stages. The partial coding sequence for a KDEL-CysEP was isolated, and its expression was determined by qRT-PCR. The gene expression did not correlate with the cysteine endoprotease activity especially in May natural flowering and November alternative forcing at 15°C. Alternative forcing method at 15°C affected the flowering process delaying senescence, presumably due to the low cysteine protease activity.
