Enhanced Expression and Primary Purification of Soluble HBD3 Fusion Protein in Escherichia coli

Abstract: To obtain in-depth information on the overall metabolic behavior of the new good xylitol producer Debaryomyces hansenii UFV-170, batch bioconversions were carried out using semisynthetic media with compositions simulating those of typical acidic hemicellulose hydrolysates of sugarcane bagasse. For this purpose, we used media containing glucose (4.3-6.5 g/L), xylose (60.1-92.1 g/L), or arabinose (5.9-9.2 g/L), or binary or ternary mixtures of them in either the presence or absence of typical inhibitors of acidi… Show more

“…The foreign protein could not be expressed fully when the IPTG concentration was too low, also the high IPTG concentration would be harmful to cell growth [18]. In this work, IPTG concentration was examined from 0.1 to 1.0 mM.…”

“…They are classified as α-, β- and θ-defensins [10] where the first two are the most common human antimicrobial peptides. The α-defensins found in neutrophils (polymorphonuclear cells, PMNs) are called human neutrophil peptides (HNP)-1, -2, -3 and -4 [11].…”

“…Hence, HBD3 protein maintaining a natural physiological concentration which is not efficiently to resist the high-density bacteria invasion within a short time. Prokaryotic expression of HBD3 fusion protein was initially designed and optimized by Huang et al ( 2006 , 2007 ). However, the existence of His tag reduced the production of HBD3.…”
