The lipophilicity of a peptide drug can be considerably increased by hydrophobic ion pairing with amphiphilic counterions for successful incorporation into lipid-based formulations. Herein, to enhance the oral absorption of insulin (INS), a self-microemulsifying drug delivery system (SMEDDS) formulation was developed. Prior to optimization, INS was complexed with sodium n -octadecyl sulfate (SOS) to increase the loading into the SMEDDS. The INS–SOS complex was characterized via scanning electron microscopy, Fourier transform infrared spectroscopy, differential scanning calorimetry, and its dissociation behavior. The SMEDDS was optimized using a D-optimal mixture design with three independent variables including Capmul MCM ( X 1 , 9.31%), Labrasol ( X 2 , 49.77%), and Tetraglycol ( X 3 , 40.92%) and three response variables including droplet size ( Y 1 , 115.2 nm), INS stability ( Y 2 , 46.75%), and INS leakage ( Y 3 , 17.67%). The desirability function was 0.766, indicating excellent agreement between the predicted and experimental values. The stability of INS-SOS against gastrointestinal enzymes was noticeably improved in the SMEDDS, and the majority of INS remained in oil droplets during release. Following oral administration in diabetic rats, the optimized SMEDDS resulted in pharmacological availabilities of 3.23% (50 IU/kg) and 2.13% (100 IU/kg). Thus, the optimized SMEDDS is a good candidate for the practical development of oral delivery of peptide drugs such as INS.

The impact of pH, temperature, and metal ions on the rheological and interfacial properties of aqueous mixed surfactant solutions composed of anionic NaC (sodium cholate) and nonionic BrijL4 [polyoxyethylene (4) lauryl ether] surfactants has been investigated. The various compound systems were analyzed, considering variations in each selected factor. The results highlight the unique characteristics of the BrijL4/NaC mixture, suggesting its potential as a viable alternative to other existing surfactants. The synergistic effect between BrijL4 and NaC significantly reduces the critical micelle concentration (CMC) and improves the wetting properties on hydrophobic surfaces, surpassing those of singlecomponent solutions. Additionally, sodium, calcium, and magnesium ions enhance surface wetting and decrease the CMC. Besides, the BrijL4/NaC solutions exhibit viscoelastic fluid behavior at higher surfactant concentrations. These viscoelastic BrijL4/NaC solutions demonstrate stability over various pH and temperature variations, exhibiting lower flow activation and scission energy values than those of other viscoelastic surfactant solutions. Notably, the BrijL4/NaC mixture has potential applications in gel-based foliar fertilizers and drug delivery systems. Furthermore, the rheological studies examine the impact of humic acid on the rheological properties of BrijL4/NaC mixture solutions, revealing that incorporating additional humic acids can achieve stable rheological properties.

Peptic ulcer disease (PUD) adversely affects ≈4 million individuals globally and remains pervasive among 5–10% of the general population. Mucosal lesions prompted by the ulceration lead to the penetration through mucosal layers, ultimately resulting in cavity formation and inflammation. Conventional therapeutic modalities for peptic ulcers have shown unfavorable effects, relapses, and numerous drug interactions. The genesis of gastric ulcers is catalyzed via an imbalanced interplay between environmental lesions, defensive mechanisms, and the restoration of gastric mucosa. Lipid based nanotechnology involved in self‐nanoemulsifying drug delivery system (SNEDDS) is being tremendously involved in opening avenues in innovative pharmaceutical delivery systems in terms of improving solubility and regenerative healing against peptic ulcers. Biological macromolecules can be incorporated into SNEDDS to embellish the drug delivery system on an advanced level. Herein, in this review, the potential of SNEDDS as a viable carrier for administering biomacromolecules for regenerative healing in ulcers is discussed in detail.
