Enhanced cleavage of type II collagen by collagenases in osteoarthritic articular cartilage.

Abstract: We demonstrate the direct involvement of increased collagenase activity in the cleavage of type II collagen in osteoarthritic human femoral condylar cartilage by developing and using antibodies reactive to carboxy-terminal (COL2-3/ 4C short ) and amino-terminal (COL2-1/4N1) neoepitopes generated by cleavage of native human type II collagen by collagenase matrix metalloproteinase (MMP)-1 (collagenase-1), MMP-8 (collagenase-2), and MMP-13 (collagenase-3). A secondary cleavage followed the initial cleavage produc… Show more

“…MMP‐3 was selected as it degrades other minor proteins in tendon matrix, including collagens III, IV, IX, and X, and also proteoglycans and elastin 28, 29, 30, 31, 32, 33. The C1,2C (or COL 2 3/4C short) antibody detects the carboxy terminus of fragmented type I and II collagen, cleaved by collagenases MMP‐1, MMP‐8 or MMP‐13 21, 34, 35, 36, 37…”

“…Many cartilage explant studies have shown that aggrecanolysis preceeds collagenolysis (Billinghurst, et al, 1997;Kuroki, et al, 2005;Pratta, et al, 2003b;Zwerina, et al, 2004), and it has been suggested (Pratta, et al, 2003b) that aggrecan protects the collagen network from proteolytic attack.…”

“…MMPs are considered to be important in the chondrolytic processes that contribute to the degenerative changes in OA cartilage (2)(3)(4). Recent studies have identified the messenger RNA (mRNA) for some MMPs, such as MMP-1, MMP-3, MMP-9, and MMP-13, in human OA cartilage (4,5), and other investigators have reported specific MMP proteins and collagenasemediated type II collagen degradation products (6,7). There is a consensus that these enzymes play a critical role in intrinsic chondrocyte-mediated degenerative changes of the cartilage matrix in OA.…”
