Engineering Zymomonas mobilis for the Production of Xylonic Acid from Sugarcane Bagasse Hydrolysate

Abstract: Sugarcane bagasse is an agricultural residue rich in xylose, which may be used as a feedstock for the production of high-value-added chemicals, such as xylonic acid, an organic acid listed as one of the top 30 value-added chemicals on a NREL report. Here, Zymomonas mobilis was engineered for the first time to produce xylonic acid from sugarcane bagasse hydrolysate. Seven coding genes for xylose dehydrogenase (XDH) were tested. The expression of XDH gene from Paraburkholderia xenovorans allowed the highest prod… Show more

“…The broad host range vector pBBR1MCS2 (Kovach et al., 1995 ) is frequently utilized in Z. mobilis (Braga et al., 2022 ; Herrera et al., 2021 ; Liu et al., 2020 ). pGB‐MPI‐068 carries the rep , oriV and mob elements of pBBR1MCS2 and was further modified to create acceptors fitting the Zymo‐Parts system.…”

“…Electrocompetent Z. mobilis ZM4 was prepared as described by Benoliel et al (2020) and Herrera et al (2021) with slight modifications. Briefly, cells were inoculated in 100 mL of RM with an optical density at 600 nm (OD 600 ) of 0.1 and incubated at 30 °C, statically (Hu et al 2021).…”

“…When it comes to other strains, the XA production was even more significantly reduced. According to a recent research, Herrera et al obtained 11.1 g/L XA from sugarcane bagasse hydrolysate with a productivity of 0.32 g/L/h using an engineered Zymomonas mobilis (Herrera et al, 2021). Thus, 335 resin detoxification appears to be a feasible method for improving the recycling-cell catalysis and bioproduction of XA from AHC.…”
