Engineering islet for improved performance by optimized reaggregation in alginate gel beads

Abstract: After islet isolation, diffusion has become the main mechanism to transport oxygen and nutrients into the core of islets. However, diffusion has limitations, by which nutrients cannot effectively reach the core of large islets and can eventually cause core cell death and islet loss. This problem can be resolved by dispersing islets into single islet cells, but single islet cells do not exhibit insulin release function in in vitro culture. In this study, we intended to establish a new islet engineering approach… Show more

“…This is of particular relevance because it has long been thought that size is a critical component to islet function, and pseudoislet dimensions must be comparable to intact islets for proper hormone secretion (14,1618,22,25,38). Although large pseudoislets have been used in transplantation and β-cell mass replacement studies, this model can also be used to interrogate the function of the other islet cell types.…”

“…Alginate as a 3D culture platform was, for instance, used for reaggregation of dispersed single islet cells with the intention of creating similar-sized islets, in order to overcome the problem of oxygen diffusion due to the wide size range in human islets. 36 Collagen is the main structural protein for most tissues and has often been investigated due to its good mechanical properties. 37 Many biological scaffold materials incorporating collagen are biologically active and show therefore good cell adhesion and proliferation properties.…”
