BackgroundThe aim of this work was to establish a process for the heterotrophic growth of green microalgae using forest biomass hydrolysates. To provide a carbon source for the growth of the green microalgae, two forest biomasses (Norway spruce and silver birch) were pretreated with a hybrid organosolv-steam explosion method, resulting in inhibitor-free pretreated solids with a high cellulose content of 77.9% w/w (birch) and 72% w/w (spruce). Pretreated solids were hydrolyzed using commercial cellulolytic enzymes to produce hydrolysate for the culture of algae.ResultsThe heterotrophic growth of A. protothecoides was assessed using synthetic medium with glucose as carbon source, where the effect of sugar concentration and the carbon-to-nitrogen ratio were optimized, resulting in accumulation of lipids at 5.42 ± 0.32 g/L (64.52 ± 0.53% lipid content) after 5 days of culture on glucose at 20 g/L. The use of birch and spruce hydrolysates was favorable for the growth and lipid accumulation of the algae, resulting in lipid production of 5.65 ± 0.21 g/L (66 ± 0.33% lipid content) and 5.28 ± 0.17 g/L (63.08 ± 0.71% lipid content) when grown on birch and spruce, respectively, after only 120 h of cultivation.ConclusionsTo the best of our knowledge, this is the first report of using organosolv pretreated wood biomass hydrolysates for the growth and lipid production of microalgae in the literature. The pretreatment process used in this study provided high saccharification of biomass without the presence of inhibitors. Moreover, the lipid profile of this microalga showed similar contents to vegetable oils which improve the biodiesel properties.

Transportation is one of the main enablers of development. It connects communities, allows the trade of goods among territories, and provides equitable access to public services. However, it is also one of the main causes of climate change, impacting directly and indirectly on human health. This raises the need for transportation infrastructure to be more sustainable so that it can enable more sustainable development. This paper investigates how sustainability in the transport sector can be conceptualized, assessed, and implemented. Definitions are proposed, indicators identified, existing methods reviewed, and three case studies presented. The Sustainable Asset Valuation (SAVi) method and resulting integrated simulation models are used to assess the multidimensional outcomes of conventional and sustainable transport investments. The cases of roads, public bicycle-sharing and bus rapid transport systems are discussed. These investments are assessed using an integrated cost–benefit analysis that goes beyond traditional assessments, considering the benefits and avoided costs of the externalities of a project in the social, environmental, and economic dimensions. The use of the method led to the conclusion that including externalities in a cost–benefit analysis is meaningful and can be relevant for assessing the economic viability of transport infrastructure projects.

Electrification of public utility vehicles plays a vital role in the transition towards a more sustainable transport system. However, the adoption of electric vehicles (EVs) encounters varying challenges ranging from financing issues, government policies, and public acceptance. Using the Philippines as a case, this research applies political, economic, social, technological, legal, and environmental (PESTLE) analysis to determine how different drivers affect the adoption of EVs in the public transport system from various transport stakeholders’ vantage points. Survey results identified economic and technological factors as the main barriers to the adoption of electric public transport. This includes high investment and operational costs, lack of charging infrastructure, issues in driving range and use in different terrains, and the availability of EV parts and repair stations. On the other hand, the main enabler is the significant public support for the modernization of the public transport system through EVs, backed up by policy and legal drivers. For a zero-emission public transport system, this study recommends that the government should invest in sustainable sources of energy, develop more public infrastructure, diversify the transport sector, fund the development of locally made EVs, and initiate a massive information campaign in educating the public of its advantages.
