When studying the steroids in the urine it must be clearly understood that, as a rule, it is not the original, physiologically active steroids that are found in the urine but their metabolites.Until just recently we have not had methods permitting a study of the minute quantities of steroids that circulate in the blood. Therefore, attention has been concentrated on their metabolites in the urine when it was a matter of clinical investigations in man. Biologic methods were used at first. The androgenic effect was determined, for example, through the growth of the comb in capons (62). The estrogenic effect can be determined by the study of vaginal smears from mice ( I ) , and different methods are available for determination of the steroids of the adrenal cortex, depending upon the aspect to be investigated. The most common is probably measurement of the glycogen deposition in the liver in adrenalectomized mice (122). Since these methods are all extremely timeconsuming and expensive and measure only the small quantities of the steroids that have biologic effect, it was a great advance when it became possible to use chemical methods. Zimmermann's discovery that m-dinitrobenzene can give a typical color reaction with the 17-ketosteroids provided the first possibility for the application of such methods (1 34). The development of different chromatographic methods for the separation of the various steroids was also of great importance (33,35, 37,101,103). Their identification has been greatly facilitated through the different spectrographic methods developed by /ones, Dobriner and Zuffaroni, inter abos (38,80,132,133).In the investigations concerning steroid metabolism and steroid excretion performed in King Gustaf V Research Institute (5-19, 63, 112) thc following methods have been used.
