Endothelin receptors mediating functional responses in human small arteries and veins

Abstract: 1 In the present study, responses of human omental small arteries and veins to endothelin-1 and endothelin-3 were characterized by use of the ETB receptor selective agonist, sarafotoxin S6c, the ETA receptor antagonist, BQ123, the ETB receptor antagonist, IRL1038, the NO-synthase inhibitor NGmonomethyl-L-arginine (L-NMMA, 300ELM) and indomethacin (10I1M).2 Small arteries (internal diameter 413 ± 22 Am) and parallel running veins (646 ± 35 Am) were mounted in a myograph under a normalized tension equivalent to … Show more

“…However, microautoradiographical visualisation of sections of human kidney using the ETA-selective [ (Karet et al, 1993). Whilst data obtained for one vascular bed cannot be extrapolated to another, it is interesting that experiments carried out using human small resistance arteries and veins in vitro suggest that endothelininduced vasoconstriction in the resistance vessels of the heart (Godfraind, 1993) and omentum (Riezebos et al, 1994), as in the larger human blood vessels, is mediated via the ETA receptor.…”

“…Only ETA receptors were apparent on the intrarenal arterioles of human kidney viewed by microautoradiography (Karet et al, 1993). Additionally, the ETA subtype has been demonstrated to mediate ET-1-induced constriction of small diameter, distal segments of human coronary artery (Godfraind, 1993) and human small omental vessels (Riezebos et al, 1994 (Tables 2 and 3). A similar difference has been observed previously in other studies with human tissue Peter & Davenport, 1995).…”

“…ET-1 exerts a variety of effects via the activation of specific receptors. Type A receptors are expressed by vascular smooth muscle cells and cause vasoconstriction (31). Type B receptors have dual functions depending on their location: when expressed by vascular smooth muscle cells, they mediate vasoconstriction, but when expressed by endothelial cells, they are linked to vasodilation via NO and/or prostacyclin synthesis (14,41).…”
