Endothelin-1 Inhibits Endothelin-Converting Enzyme-1 Expression in Cultured Rat Pulmonary Endothelial Cells

Abstract: Background-The lung expresses large amounts of endothelin-converting enzyme-1 (ECE-1), which catalyzes a step in the biosynthesis of potent vasoactive endothelin-1 (ET-1) from the inactive intermediate big ET-1. Because there has been no report concerning a possible relationship between ET-1 and ECE-1, we investigated the effects of ET-1 on ECE-1 expression in cultured rat pulmonary endothelial cells. Methods and Results-ECE-1 messenger RNA (mRNA) and protein expression in cultured endothelial cells were assay… Show more

“…Increased plasma ET-1 has also been reported in patients with PAH [6, 36, 37], though this may reflect increased production rather than impaired pulmonary clearance [38]. Loss of ET B signalling has also been reported to increase ECE-1 mRNA expression [39], which may further contribute to an increase in ET-1. However, the limited experimental evidence available suggests that an isolated increase in ET-1 is insufficient to cause PAH.…”

“…ECE-1 expression is regulated by protein kinase C (PKC), ET B receptors, the transcription factor ets-1, and cytokines [10,[23][24][25][26]. ECE-1 is found in a variety of cells including endothelial cells, has peak activity at neutral pH, and is processed both intracellularly and on the cell surface [9,27].…”

“…A vascular endothelial cell line originally established from rat lung (TRLEC) (17,18) was kindly donated by the Institute of Cytosignal Research Inc. (Tokyo, Japan), and was cultured in DMEM containing 10% FBS. Human adult dermal microvascular endothelial cells (hADM) and human aortic smooth muscle cells (hASM) were purchased from Cell Systems (Kirkland, USA), and were cultured in a half-Nutrient Mixture F-12 Ham/half-DMEM mixture containing endothelial cell growth supplement, heparin, glutamine, and 10% FBS.…”
