Endothelial Wnts regulate β‐catenin signaling in murine liver zonation and regeneration: A sequel to the Wnt–Wnt situation

Abstract: β‐Catenin in hepatocytes, under the control of Wnts, regulates pericentral gene expression. It also contributes to liver regeneration (LR) after partial hepatectomy (PH) by regulating cyclin‐D1 gene expression as shown in the β‐catenin and Wnt coreceptors low‐density lipoprotein receptor‐related protein 5/6 conditional knockouts (KO). However, conditional deletion of Wntless (Wls), required for Wnt secretion, in hepatocytes, cholangiocytes, or macrophages lacked any impact on zonation, while Wls deletion in ma… Show more

“…Wnt is a major factor that shapes hepatocyte zonation [35][36][37][38][39][40] . Wnt and Rspondin morphogens are secreted by pericentral liver endothelial cells 2,11,[41][42][43] , resulting in higher pericentral expression of Wnt-activated genes and lower pericentral expression of Wnt-inhibited genes 5,35 . Notably, hepatocyte-specific Dicer knock-out mice have perturbed zonation of Wnt-regulated genes, such as Glul and Arg1 24 .…”

“…It is acknowledged that hepatic parenchymal turnover is maintained solely through hepatocyte division controlled by some pivotal signaling pathways, including Wnt/ b-catenin and Hippo/Yap [6,7]. This spatiotemporal regulation contributes to metabolic zonation and determines hepatocyte function [8]. Rodent models such as the classical partial hepatectomy in rats demonstrate that the liver size is restored through hepatocyte hypertrophy and hyperplasia in the remaining lobes.…”

“…110 Hepatic macrophages isolated from APAP-injured mouse liver stimulated LSEC proliferation and migration in vitro suggesting paracrine proangiogenic function. 105 Macrophages are also known to express several WNT ligands, 111,112 which are known to stimulate β-catenin signaling in hepatocytes during liver regeneration. 113 CSF1R-mediated hepatic macrophage accumulation induced a modest but significant increase in hepatocyte proliferation after partial hepatectomy suggesting macrophages may promote parenchymal cell division, although specific factors underpinning this have not been identified.…”
