Endothelial Transcytosis of Lipoproteins in Atherosclerosis

Abstract: Seminal studies from Nikolai Anichckov identified the accumulation of cholesterol in the arteries as the initial event that lead to the formation of atherosclerotic plaques. Further studies by Gofman and colleagues demonstrated that high levels of circulating low-density lipoprotein cholesterol (LDL-C) was responsible for the accelerated atherosclerosis observed in humans. These findings were confirmed by numerous epidemiological studies which identified elevated LDL-C levels as a major risk factor for cardiov… Show more

“…Generally, the LDL-C enters the intimal layer by the mechanism of transcytosis by sieving through the endothelial cells. 27 The translocated LDL undergoes the following changes: oxidation, acylation, lipolysis, proteolysis, aggregation, and fusion in the intima. 28 The initial product of oxidation of LDL is the minimally OxLDL, which further oxidized to form OxLDL.…”

“…Upon internalisation, the cargo in the endocytic vesicle enters a complex trafficking pathway. Some vesicles are transported across the cell (transcytosis); this occurs predominantly after caveolin‐mediated endocytosis [16,20,21]. Most endocytic vesicles are quickly targeted to the early endosome (EE) (Figure 3), a sorting station, where the fate of the cargo is determined.…”

“…Due to its tight specialized cell-to-cell junctions, the endothelium forms a barrier that selectively limits the movement of macromolecules (Rahimi, 2017). The barrier is critically involved in vascular tone, fluid homeostasis, and host defense (Zhang et al, 2018b). Activated endothelial cells may release various cytokines, chemokines, and growth factors that promote the proliferation, migration, and permeability of endothelial cells (Park-Windhol and D'amore, 2016).…”
