Endothelial Nitric Oxide Synthase Haplotypes Associated with Hypertension Do Not Predispose to Cardiac Hypertrophy

Abstract: Left ventricular hypertrophy (LVH) is a complication that may result from chronic hypertension. While nitric oxide (NO) deficiency has been associated with LVH, inconsistent results have been reported with regards to the association of endothelial NO synthase (eNOS) polymorphisms and LVH in hypertensive patients. This study aims to assess whether eNOS haplotypes are associated with LVH in hypertensive patients. This study included 101 healthy controls and 173 hypertensive patients submitted to echocardiography… Show more

“…Preparation of the reactions was performed according to the manufacturer's specifications for each sample: 1x of the master mix, 1x of TaqMan genotyping assay, and 50 ng of template DNA in 10 μ l final volume. Real-time PCR was performed on StepOnePlus equipment (Applied Biosystems, USA) and analyzed with the manufacturer's software [ 35 ].…”

“…Nitric oxide (NO) generated from endothelial NO synthase (eNOS) is the critical biochemical that mediates the response to shear stress in cell culture and animal studies [8,9]. This is clinically supported by the association of hypertension and eNOS haplotypes in human studies [10]. In line with human data, chronic NOS inhibition in rat or eNOS gene deletion in mice results in significant blood pressure elevation [11,12].…”

“…The reaction was carried out using Custom Taqman allele discrimination assay (re-synthesis part number AH5I790, Thermo Fisher, United States) and JumpStart Ready Mix for Quantitative PCR (Sigma-Aldrich, United States). Preparation of the reactions was performed according to the manufacturer's specifications (Vasconcellos et al, 2010). The NOS3 SNP at the position 894G > T (Glu298Asp) (rs1799983) was analyzed by conventional PCR followed by electrophoresis, using the following primers-forward: 5 -CATGAGGCTCAG CCCCAGAAC-3 ; reverse: 5 -AGTCAATCCCTTTGG TGC TCAC-3 .…”
