Endothelial microparticle-associated protein disulfide isomerase increases platelet activation in diabetic coronary heart disease

Abstract: Background: Endothelial microparticles (EMPs) carrying the protein disulfide isomerase (PDI) might play a key role in promoting platelet activation in diabetes. This study aimed to examine the activation of platelets, the amounts of MPs, PMPs, and EMPs, and the concentration and activity of PDI in patients with diabetic coronary heart disease (CHD) and non-diabetic CHD. Methods: Patients with CHD (n=223) were divided as non-diabetic CHD (n=121) and diabetic CHD (n=102). Platelet activation biomarker… Show more

“…As promising vesicular markers associated with endothelial dysfunction in Type 2 diabetes mellitus (DM2) are the level of CD31+CD144+ EVs, the level of miR-126 in circulating EVs, and an increased ratio of CD31+Annexin V+ EVs to CD31+CD62+ EVs, reflecting the altered immune phenotype of endothelial cell-derived EVs [ 3 , 4 ]. Vesicular markers are successfully used as markers of an increased risk of cardiovascular disease complications in obesity, DM2, and metabolic syndrome (perilipin A, cystatin C, CD14, miR21, disulfide isomerase levels on CD31-positive BBs) [ 3 , 5 , 6 , 7 ]. Several miRNAs have been identified in urinary EVs associated with diabetic nephropathy and renal fibrosis.…”

“… 10 Furthermore, in patients with diabetes with CVD, the PDMPs were multiplied, indicating platelet activation in this population. 11 During an AMI episode, more enhanced platelet activation and subsequent platelet-enriched thrombus formation among patients with diabetes mellitus may amplify the production of PDMPs. However, despite the abundant data on increased circulating PDMPs among diabetes mellitus and stable CVD, the impact of acute hyperglycemia and diabetes mellitus during AMI on circulating PDMPs release has not yet been investigated.…”
