Endothelial Cell Plasma Membrane Biomechanics Mediates Effects of Pro-Inflammatory Factors on Endothelial Mechanosensors: Vicious Circle Formation in Atherogenic Inflammation

Abstract: Chronic low-grade vascular inflammation and endothelial dysfunction significantly contribute to the pathogenesis of cardiovascular diseases. In endothelial cells (ECs), anti-inflammatory or pro-inflammatory signaling can be induced by different patterns of the fluid shear stress (SS) exerted by blood flow on ECs. Laminar blood flow with high magnitude is anti-inflammatory, while disturbed flow and laminar flow with low magnitude is pro-inflammatory. Endothelial mechanosensors are the key upstream signaling pro… Show more

“…The glycocalyx is a dense macromolecular mesh composed of proteoglycans and glycosaminoglycans decorating the endothelial outer membrane, and has been shown to be both a primary mechanosensor and final effector for cellular adaptation to FSS ( Pahakis et al, 2007 ; Psefteli et al, 2021 ; Wang et al, 2020 ; Zeng and Tarbell, 2014 ; Zeng et al, 2018 ; Barvitenko et al, 2022 ) ( Fig. 2 ).…”

“…The balance of anti- and proinflammatory factors determines the trend of the inflammatory response. Anti-inflammatory factors predominantly include IL-4/10/35 and transforming growth factor β (TGF-β); proinflammatory factors mainly include IL-1β/2/6/15/16/17; tumour necrosis factor (TNF); and interferon γ (IFN-γ) [ 98 100 ] . In this section, the production and secretion mechanisms of inflammatory factors in the SASP are discussed, and the epistemic regulatory mechanisms of the SASP are revealed, providing new ideas for the prevention and treatment of aging.…”

“…The review of Nadezhda Barvitenko et al [ 14 ] indicates that endothelial mechanosensors are the key upstream signaling proteins in shear stress (SS)-induced pro- and anti-inflammatory responses. As transmembrane proteins, mechanosensors not only experience fluid SS but also become regulated by the biomechanical properties of the lipid bilayer and the cytoskeleton.…”
