It has largely been responsible for the "green revolution", i.e. the massive increase in food production obtained from the same surface of land with the help of mineral fertilisers (nitrogen, phosphorus, potassium), more efficient machinery and intensive irrigation. The use of pesticides helped to significantly reduce crop losses and to improve the yield of crops such as corn, maize, vegetables, potatoes and cotton. Notwithstanding the beneficial effects of pesticides, their adverse effects on environmental quality and human health have been well documented worldwide and constitute a major issue that gives rise to concerns at local, regional, national and global scales (Planas et al., 1997; Huber et al., 2000; Kidd et al., 2001; Cerejeira et al., 2003). Residues of pesticides contaminate soils and water, persist in the crops, enter the food chain, and finally are ingested by humans with foodstuffs and water. Furthermore, pesticides can be held responsible for contributing to biodiversity losses and deterioration of natural habitats (Sattler et al., 2006). There have been reported instances of pest resurgence, development of resistance to pesticides, secondary pest outbreaks and destruction of non-target species. Palabras clave: plaguicidas orgánicos persistentes, extracción y limpieza, metodología analítica RESUMEN Se revisaron las técnicas de extracción, limpieza y cuantificación de endosulfán por métodos cromatográficos en matrices alimentarias como productos lácteos, frutas y vegetales, así como pescados y mariscos, publicados en los últimos veinte años. Independientemente del tipo de matriz, su extracción se realiza principalmente con disolventes orgánicos como hexano, metanol, acetonitrilo y acetona. Para la limpieza, lo más comúnmente empleado fueron cartuchos de sílice, columnas de florisil, o C18. En cuanto a la cuantificación, el método más sensible resultó ser la cromatografía de gases acoplada a espectrofotometría de masas (CG-MS). Los alimentos en los que se reportó la presencia de α, β y/o sulfato de endosulfán fueron: productos lácteos y sus derivados con un intervalo de concentraciones de 23.99 a 52.2 ng/g; pescados y mariscos de 1.07 a 49.7 ng/g; y vegetales de 0.5 a 5460.0 ng/g. En leche materna se reportaron concentraciones de 0.4 a 10.7 ng/mL y en grasa hasta 277.39 ng/g. Keywords: persistent organic pesticides, extraction and cleaning, analytic methodology ABSTRACT Extraction and cleaning techniques plus chromatographic methods were reviewed for the quantification of endosulfan in food matrices such as dairy products, fruits and vegetables, fish and seafood, published in the last twenty years. Regardless of the type of matrix, the extraction of endosulfan is carried out mainly with organic solvents such as hexane, methanol, acetonitrile and acetone. For cleaning, the most common was the use silica cartridges, florisil columns, or C18. The most sensitive quantification method was gas chromatography coupled to mass spectrometry (GC-MS. Foods where it was reported the presence of α, β and/or endosulfan sulfate were dairy products and their derivatives in a range of 23.99 to 52.2 ng/g, in fish and shellfish from 1.07 to 49.7 ng/g, vegetables from 0.5 to 5460.0 ng/g. BACKGROUND: Endosulfan has been used for over 50 years. Although most analogs have been discontinued, endosulfan has less environmental persistence. Nevertheless, pressure groups are lobbying for a worldwide ban. The reasons are: possible rodent male reproductive toxicity, other endocrine effects and cancer; human epidemiology, and exposure studies; residues appearing in remote areas of the world, e.g., the Arctic. METHODS: The endosulfan toxicology database is described and risks of its use assessed. RESULTS: Endosulfan is an antagonist at the GABA A receptor Cl À ionophore in mammalian CNS. Rat acute toxicity is moderate, LD 50 5 48 (M) or 10 mg/kg/d (F), oral gavage; 130 (M), 70 mg/kg/d (F) dermal; LC 50 5 34.5 mg/L (M), 12.6 mg/L (F), inhalation. Critical NOELs for risk assessment: acute oral (gavage) 5 0.7 mg/kg/d (rabbit developmental); Subchronic oral (diet) 5 1.2 mg/kg/d (rat reproduction); Chronic oral (diet) 5 0.6 mg/kg/d. There were no acceptable dermal toxicity studies. The critical acute and subchronic inhalation NOELs 5 0.001 mg/L, chronic inhalation 5 0.0001 mg/L (estimated). Toxicity to rat sperm occurred at doses causing neurotoxicity. Endocrine effects, resulting from P450 oxygenase(s) induction, were reversible. Increased cancer, genotoxicity, or histopathology in rodents was not observed in any organ. Possible effects on brain biogenic amine levels were probably secondary. CONCLUSIONS: Epidemiology and rodent studies suggesting autism and male reproductive toxicity are open to other interpretations. Developmental/ reproductive toxicity or endocrine disruption occurs only at doses causing neurotoxicity. Toxicity to the fetus or young animals is not more severe than that shown by adults.
