Endostatin attenuates PDGF-BB- or TGF-β1-induced HSCs activation via suppressing RhoA/ROCK1 signal pathways

Abstract: AimTo testify the hypothesis that endostatin exerts antifibrotic effects in hepatic stellate cells (HSCs) by modulating RhoA (ras homolog gene family, member A)/ROCK 1 (Rho-associated protein kinase 1) signal pathways.Materials and methodsHSCs-T6 of passages 3–5 were cultured in DMEM and serum starved for 48 hours. HSCs were grouped as follows: control group, TGF-β1 (transforming growth factor β1) group, endostatin+TGF-β1 group, PDGF-BB (platelet-derived growth factor-BB) group, and endostatin+PDGF-BB group. “…The increased type XVIII collagen upregulates its own product, EST. EST is able to resist liver fibrosis by inhibiting the expression of TGF‐β1 mRNA through RhoA/ROCK signal pathways in hepatic stellate cells (HSCs) 47,48 . In addition, liver fibrosis refers to the progression of extracellular matrix excessive deposition, which is often promoted by the activation of HSCs.…”

“…It is also involved in the development of liver diseases (i.e., nonalcoholic steatohepatitis and hepatocellular carcinoma) [409], whereas endostatin is antifibrotic [410]. Canstatin is anti‐lymphangiogenic and modulates voltage‐dependent calcium channel activity in rat cardiomyocytes [411], and endostatin‐derived peptides are antifibrotic [410]. ECM bioactive (matricryptins and matrikines) have potential applications in therapeutics, tissue engineering [402,412], and cosmetics [413].…”

“…Endostatin is reported to inhibit the RhoA/ROCK pathway. Recent findings confirm that endostatin suppresses HSCs activation by inhibiting RhoA/ROCK1 signaling pathway (Ren et al 2019). HSCs can be triggered by factors like inflammation or injury.…”
