Sucretolerant microbes grow in the presence of sugar concentrations high enough to substantially lower water activities. Natural habitats high in sugars are mainly limited to dried fruit, floral nectar, honey, sugarcane, and associated soils. Organisms that tolerate extremes of solute concentration, high enough to lower water activities, might not be expected in common oligoosmotic soils. We report on the isolation of sucretolerant bacteria from common soils using media supplemented with 50% sucrose (aw 0.91) and their physiological characterization and identification by 16S rRNA gene phylogeny. Fifteen of these sucretolerant isolates from common soils were related to four Bacillus spp. A Lysinibacillus and a Microbacterium (actinomycete) also were collected. All grew at 50% sucrose and 13 grew at 60% sucrose. Most probable number counts were used to determine the abundance of sucretolerant microbes in several common soil types, including agricultural, managed turf, and native prairie. Microbial abundance (with fungicides) was about 105 and 103 cells g−1 soil in media containing 50 or 70% sucrose, respectively. The abundances of sucretolerant bacteria in common soils mirror those of halotolerant bacteria that grow at 10 and 20% NaCl. However, there is not a correlation between halotolerance and sucretolerance in our isolates, nor can predictions be made based on taxonomy. Specific solute effects may be at work, rather than biological responses to a single physicochemical parameter such as aw. The occurrence of spore-forming sucretolerant bacteria in common soils has relevance to forward planetary protection and astrobiology. Extraterrestrial habitable regions are defined in part by tolerance to high solute concentrations and osmotolerant soil microbes may contaminate spacecraft.

Airborne bacteria are dispersed along with Asian dust and have various influences on atmospheric characteristics, downwind ecosystems, and public health. In this study, aircraft observations were conducted during Asian dust passage and non-dust conditions over the East China Sea in December 2010, and the composition of culturable bacteria within the dust was investigated using culture-based gene analysis. The 16S rRNA gene analysis revealed that the 34 cultured strains isolated from the transported dust could be grouped into Bacillales and Actinomycetales. Highly tolerant endospore-forming bacteria (25 isolates) were predominant in the composition. On the other hand, all non-endospore-forming bacteria (9 isolates) were gram-positive bacteria with high guanine and cytosine contents, which have substantial ultraviolet (UV) resistance. Thus, the isolated culturable bacteria were strains having some tolerance for exposure to UV radiation. Although the atmosphere is an extreme environment for bacterial survival, Asian dust can facilitate atmospheric dispersion of culturable bacteria that are resistant to the harsh ambient air environment.

Polycyclic aromatic hydrocarbons (PAHs) are known as carcinogenic and/or mutagenic substances, and are present at high concentration in polluted environments. It has recently been reported that spore-forming bacteria (e.g., Bacillus spp.) can be transported long distances alive in the atmosphere, which raises the possibility that some of the transported bacteria could have adverse effects on human health. There is thus a need for filters that can remove gaseous PAHs from the air that people breathe and that can inhibit bacterial growth on the filters. We focused on metallophthalocyanine derivatives (M-Pc) which are known to adsorb PAHs as well as to inhibit the growth of bacteria as a potential filtering agent. In this study, we developed different types of M-Pc-supported rayon fibers by changing central metals, functional groups, concentrations of M-Pc and rayon types, and evaluated their removal effects by measuring adsorption rates of 3-and 4-ring PAHs with a HPLC and growth curves of Bacillus sp. with a spectrophotometer. The results showed that both the effects depended on functional groups and concentrations of M-Pc, and rayon types. The most effective combination was observed in Fe-Pc with sulfo group supported on cationized rayon fiber at the concentration of 2 to 3.3 wt%. Central metal species of M-Pc were influenced only on the antibacterial properties. This fiber would be applicable to filtering agents and textiles.
