Endoplasmic Reticulum–Plasma Membrane Contact Sites: Regulators, Mechanisms, and Physiological Functions

Abstract: The endoplasmic reticulum (ER) forms direct membrane contact sites with the plasma membrane (PM) in eukaryotic cells. These ER-PM contact sites play essential roles in lipid homeostasis, ion dynamics, and cell signaling, which are carried out by protein-protein or protein-lipid interactions. Distinct tethering factors dynamically control the architecture of ER-PM junctions in response to intracellular signals or external stimuli. The physiological roles of ER-PM contact sites are dependent on a variety of regu… Show more

“…PM cholesterol therefore serves as first regulator in a viral life cycle, which is either supplied by ER-PM contact sites or ERC-PM contact sites. While various adaptors for ER-PM shuttling have been identified ( Naito et al, 2019 ; Li et al, 2021 ), ERC-PM contacts present as rather enigmatic although evidently being similarly important ( Hölttä-Vuori et al, 2002 ; Möbius et al, 2003 ; Garbarino et al, 2012 ; Iaea et al, 2017 ). Further elucidation of the latter and how it is modulated by viruses thus presents as important aspect of focus.…”

“…The ER can also form Ca 2+ conducive membrane interphases with the PM (76) and this interaction is critical for the process of store operated Ca 2+ entry (SOCE) into the ER and cytosol from the extracellular space with the tethering of the membranes mediated by the proteins ORAI and Stromal interaction molecules (STIMs) (76). SOCE plays an important role in the nerve terminals where α-syn is present in high concentrations (77).…”

“…This symmetric process, on either side of the cell wall at the graft interface, would require the precise spatial-temporal coordination of processes between the scion and the rootstock. This coordination could be mediated by signals produced during the cell wall remodeling (For reviews: Anderson and Kieber, 2020; Duman et al, 2020) and/or by local ER-plasma membrane interactions (For review: Li et al, 2021a). The genesis of a large central cavity, by for example the formation of hemi-PD running in parallel to the cell wall, could increase the chance of alignment of both hemi-PD on either side of the graft interface.…”
