Fungal-bacterial combinations have a significant role in increasing and improving plant health under various stress conditions. Metabolites secreted by fungi and bacteria play an important role in this process. Our study emphasizes the significance of secondary metabolites secreted by the fungus Serendipita indica alone and by an actinobacterium Zhihengliuella sp. ISTPL4 under normal growth conditions and arsenic (As) stress condition. Here, we evaluated the arsenic tolerance ability of S. indica alone and in combination with Z. sp. ISTPL4 under in vitro conditions. The growth of S. indica and Z. sp. ISTPL4 was measured in varying concentrations of arsenic and the effect of arsenic on spore size and morphology of S. indica was determined using confocal microscopy and scanning electron microscopy. The metabolomics study indicated that S. indica alone in normal growth conditions and under As stress released pentadecanoic acid, glycerol tricaprylate, L-proline and cyclo(L-prolyl-L-valine). Similarly, d-Ribose, 2-deoxy-bis(thioheptyl)-dithioacetal were secreted by a combination of S. indica and Z. sp. ISTPL4. Confocal studies revealed that spore size of S. indica decreased by 18% at 1.9 mM and by 15% when in combination with Z. sp. ISTPL4 at a 2.4 mM concentration of As. Arsenic above this concentration resulted in spore degeneration and hyphae fragmentation. Scanning electron microscopy (SEM) results indicated an increased spore size of S. indica in the presence of Z. sp. ISTPL4 (18 ± 0.75 µm) compared to S. indica alone (14 ± 0.24 µm) under normal growth conditions. Our study concluded that the suggested combination of microbial consortium can be used to increase sustainable agriculture by combating biotic as well as abiotic stress. This is because the metabolites released by the microbial combination display antifungal and antibacterial properties. The metabolites, besides evading stress, also confer other survival strategies. Therefore, the choice of consortia and combination partners is important and can help in developing strategies for coping with As stress.

In this study, a fungal endophyte was isolated from Anaphalis contorta, a medicinal plant and identified as Plectosphaerella oligotrophica using ITS-rDNA gene sequencing that shows 99.61% similarity. Production of hydrolytic enzymes by this fungus was determined. The isolate was allowed to biosynthesize silver nanoparticles(AgNPs)which was confirmed using UV-Vis spectrophotometer. The biological activities of P. oligotrophica and AgNPs were assessed in vitro. The antifungal activity of P. oligotrophica was evaluated using a modified dual culture method against ten phytopathogens and found to show highest activity against A. flavus, U. virens, A. tenuisimma, and no activity against A. niger. The AgNPs have shown antifungal activity against all the tested pathogens with the highest inhibition against U. virens (60%). Antibacterial activity was conducted using the agar well diffusion method and 96-well microtitre method against six pathogenic bacteria. The ethyl acetate extract of P. oligotrophica was found to possess weak to moderate antibacterial activity showing highest activity against S. typhi with the MIC of 62.50 µg/ml. The AgNPs have shown strong antibacterial activity with the greatest inhibition against S. typhi having MIC of 15.62 µg/ml. The antioxidant activity was assessed using DPPH assay. The P. oligotrophica extract and AgNPs have shown inhibitions of 49.85% and 59.56%, respectively.The biosynthesized AgNPs displayed potential biological activities and hence provided an important application for agricultural sciences, medical sciences, and pharmaceutical industries.

Endophytes are the beneficial group of plant symbionts and are potent sources for producing novel metabolites with biotechnological applications. They are efficient sources for the synthesis of plant growth-promoting substances- Indole acetic acid, ammonia, solubilizing phosphates and thus open up sustainable agricultural opportunities. In the present investigation, twenty-two fungal endophytes were isolated from an ethnomedicinal plant-Helicteres isora L., collected from forests of Singbhum, Jhargram, India, and the isolate-Colletotrichum sp. HelS1, exhibited the most potent in vitro plant growth-promoting activities among all. HelS1 synthesized auxin (111.13 µg ml-1), ammonia, and solubilized phosphate (47.22 µg ml-1) in specific culture conditions. A treatment with fungal extract effectively increases the growth of the experimental plant (tomato seedlings) in terms of improvement in relative water contents, dry weight, nodal length, and pigment profiles compared to the untreated ones. There was an increase in root length by 27% compared to the control group. The isolate produced the maximum IAA after 7 days of incubation on Czepak Dox Broth supplemented with tryptophan (5 mg mL-1), sucrose (30 g L-1), and NaNO3 (2.2 g L-1) at pH 6.2. Fourier Transform Infrared Spectroscopic analysis of the crude fungal extract further confirmed the production of indole-like compounds. This investigation suggested that secondary metabolites of endophytes act as a potent plant growth inducer and can be utilized in bio-based crop management techniques.
